【摘要】：目的探討抑制肽�；彼崦搧啺泵�4(PADI4)對卵巢癌細(xì)胞系OVCAR3的影響。方法 PADI4-si RNA組和陰性對照組si RNA分別轉(zhuǎn)染卵巢癌細(xì)胞OVCAR3,采用實(shí)時(shí)定量PCR及Western Blotting檢測PADI4、p53和p21的表達(dá),AnnexinⅤ-PE/7-AAD雙染和transw ell小室法分別檢測細(xì)胞凋亡、侵襲和遷移的情況。結(jié)果與陰性對照組相比,PADI4-si RNA組的OVCAR3細(xì)胞中PADI4表達(dá)降低,且其凋亡細(xì)胞比例明顯增加(P0.05),細(xì)胞侵襲能力明顯減弱(P0.01),但細(xì)胞趨化能力沒有明顯變化。PADI4-si RNA組p53表達(dá)較陰性對照組降低,但p21表達(dá)并沒有降低。結(jié)論沉默PADI4影響卵巢癌細(xì)胞OVCAR3凋亡和侵襲特性,提示PADI4參與調(diào)控卵巢癌細(xì)胞惡性生物學(xué)行為。
[Abstract]:Objective to investigate the inhibition of peptidyl arginine deaminase 4 (PADI4). Methods the PADI4-si RNA group and the negative control group (si RNA) were transfected into the ovarian cancer cell line OVCAR3, respectively.
