【摘要】：目的:探討CXCR4在宮頸癌細(xì)胞株Hela細(xì)胞中的表達(dá)情況,及CXCL12-CXCR4生物軸對(duì)Hela的增殖、遷移和侵襲能力的影響,并研究CXCR4的特異性拮抗劑AMD3100對(duì)CXCR4的特異性阻斷效果。方法:將宮頸癌Hela細(xì)胞株細(xì)胞進(jìn)行體外培養(yǎng)。免疫組化檢測(cè)Hela細(xì)胞中CXCR4的表達(dá)情況。采用MTT法研究不同濃度CXCL12對(duì)Hela細(xì)胞增殖的影響;Transwell法進(jìn)行實(shí)驗(yàn),判斷在無(wú)血清培養(yǎng)液的生長(zhǎng)條件下,不同濃度的CXCL12和CXCR4拮抗劑AMD3100對(duì)Hela細(xì)胞遷移、侵襲的影響。結(jié)果:CXCR4在Hela細(xì)胞的細(xì)胞膜、細(xì)胞質(zhì)中有表達(dá)。1組Hela細(xì)胞的增殖數(shù)與2組及3組兩兩比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05),4組Hela細(xì)胞的增殖數(shù)較2組、3組低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。與A組Hela細(xì)胞的遷移數(shù)和侵襲數(shù)相比,B組明顯更高(P0.05);而C組Hela細(xì)胞的遷移數(shù)和侵襲數(shù)明顯高于A組和B組;D組Hela細(xì)胞的遷移數(shù)和侵襲數(shù)均低于A、B、C3組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:CXCR4在Hela細(xì)胞的細(xì)胞膜、細(xì)胞質(zhì)中均表達(dá)。CXCL12-CXCR4生物軸可促進(jìn)宮頸癌Hela細(xì)胞株的遷移及侵襲,而CXCR4拮抗劑AMD3100能抑制這一過程,CXCL12-CXCR4生物軸在宮頸癌Hela細(xì)胞生長(zhǎng)和轉(zhuǎn)移過程中可能起著重要作用。
[Abstract]:Objective: to investigate the expression of CXCR4 in cervical cancer cell line Hela and the effect of CXCL12-CXCR4 axis on the proliferation, migration and invasion of Hela. The specific blocking effect of AMD3100, a specific antagonist of CXCR4, on CXCR4 was studied. Hela cells of cervical cancer were cultured in vitro. The expression of CXCR4 in Hela cells was detected by immunohistochemistry. The effect of different concentration of CXCL12 on Hela cells was studied by MTT method. The effects of colonization; Transwell assay was used to determine the migration of Hela cells in different concentrations of CXCL12 and CXCR4 antagonist AMD3100 under the condition of serum-free medium. Results the proliferation of Hela cells in the cell membrane and cytoplasm of Hela cells was compared with that of group 2 and group 3. The proliferative number of Hela cells in P0. 05 group was lower than that in group 2 and group 3. Compared with group A, the migration and invasion of Hela cells in group B were significantly higher than those in group B (P 0.05). The number of migration and invasion of Hela cells in group C was significantly higher than that in group A and group B. The migration and invasion of Hela cells in group D were lower than those in group C (P 0.05). The expression of. CXCL12-CXCR4 in cytoplasm can promote the migration and invasion of cervical cancer Hela cell line, but AMD3100 can inhibit this process. CXCL12-CXCR4 axis may play an important role in the growth and metastasis of cervical cancer Hela cells.
