miR-210與子癇前期滋養(yǎng)層細(xì)胞自噬的關(guān)系

【摘要】:目的探討子癇前期胎盤組織中miR-210和自噬情況,并初步闡明其可能的分子機(jī)制。方法收集子癇前期患者(子癇前期組)和同期住院正常晚期妊娠孕婦(正常對照組)各20例,應(yīng)用Real-time PCR檢測胎盤組織中miR-210和自噬標(biāo)志基因Beclin-1、LC3Ⅱ的表達(dá)水平,分離培養(yǎng)胎盤滋養(yǎng)層細(xì)胞,采用miR-210抑制劑(anti-miR-210)敲低miR-210,通過real-time PCR檢測Beclin-1、LC3Ⅱ的表達(dá)變化情況。結(jié)果子癇前期組胎盤組織內(nèi)miR-210表達(dá)增加,自噬增多,Beclin-1和LC3Ⅱ表達(dá)量較正常對照組明顯升高,敲低miR-210后顯著抑制自噬反應(yīng),Beclin-1和LC3Ⅱ的表達(dá)下降,表明二者之間存在協(xié)同關(guān)系。結(jié)論 miR-210參與調(diào)控子癇前期滋養(yǎng)層細(xì)胞自噬。
