γ-神經(jīng)突觸核蛋白基因沉默對子宮內(nèi)膜癌HEC-1A細(xì)胞周期的影響

【摘要】:目的觀察γ-神經(jīng)突觸核蛋白(SNCG)基因穩(wěn)定沉默后人子宮內(nèi)膜癌HEC-1A細(xì)胞生長周期的變化。方法采用流式細(xì)胞術(shù)檢測SNCG基因沉默后HEC-1A細(xì)胞周期變化;通過激光掃描共聚焦顯微鏡觀察吖啶橙染色后G2/M期百分比變化。結(jié)果流式細(xì)胞術(shù)結(jié)果顯示,SNCG基因沉默后HEC-1A細(xì)胞處于G_1和G_2/M期增多,處于S期減少,差異有統(tǒng)計學(xué)意義(P0.05);共聚焦顯微鏡觀察結(jié)果顯示,SNCG基因沉默后HEC-1A細(xì)胞處于G_2/M期百分比增高,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論兩個方法共同驗證了慢病毒質(zhì)粒載體shRNA干擾后SNCG基因穩(wěn)定沉默的HEC-1A細(xì)胞周期阻滯于G_2/M期,提示SNCG基因與子宮內(nèi)膜癌細(xì)胞生長周期密切相關(guān)。
[Abstract]:Objective to observe the expression of 緯 -neurosynaptophysin (SNCGG). Methods the changes of HEC-1A cell cycle after SNCG gene silencing were detected by flow cytometry. The percentage change of G _ 2 / M phase after acridine orange staining was observed by laser scanning confocal microscope. After SNCG gene silencing, the number of HEC-1A cells increased in Gsta1 and G2 / M phase, and decreased in S phase. The difference was statistically significant (P 0.05). The results of confocal microscopy showed that the percentage of HEC-1A cells in 2 / M phase was increased after the silencing of SNCG gene. The difference was statistically significant (P0.05). Conclusion the two methods combined to verify that the stable silencing HEC-1A cell cycle of SNCG gene after the interference of lentivirus plasmid shRNA was blocked in G _ S _ 2 / M phase. The results suggest that SNCG gene is closely related to the growth cycle of endometrial cancer cells.
