肝X受體α、膽固醇調(diào)節(jié)元件結(jié)合蛋白1c介導(dǎo)脂質(zhì)代謝紊亂在子癇前期中的作用及機(jī)制

【摘要】:目的研究子癇前期患者血清和胎盤(pán)中肝X受體α(LXRα)及其靶基因膽固醇調(diào)節(jié)元件結(jié)合蛋白1c(SREBP-1c)的表達(dá)與脂質(zhì)代謝紊亂的關(guān)系及機(jī)制。方法入選子癇前期患者32例(輕度子癇前期15例和重度子癇前期17例)、正常妊娠30例和正常未孕30例。測(cè)定各組血脂水平,計(jì)算動(dòng)脈硬化指數(shù)[動(dòng)脈硬化指數(shù)=(總膽固醇-高密度脂蛋白膽固醇)/高密度脂蛋白膽固醇];酶聯(lián)免疫吸附試驗(yàn)(ELISA)測(cè)定各組血清中LXRα及SREBP-1c蛋白水平;逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)及免疫組化技術(shù)(IHC)檢測(cè)胎盤(pán)中氧化型低密度脂蛋白(oxLDL)、LXRα及SREBP-1c的mRNA和(或)蛋白表達(dá)水平。結(jié)果子癇前期組與正常妊娠組比較,總膽固醇[(6.78±1.56)比(5.20±0.96)mmol/L]、三酰甘油[(3.92±1.35)比(2.48±0.78)mmol/L]、極低密度脂蛋白膽固醇[(1.85±0.53)比(1.12±0.35)mmol/L]、低密度脂蛋白膽固醇[(4.31±1.23)比(3.01±0.55)mmol/L]、動(dòng)脈硬化指數(shù)(3.10±1.22比1.88±0.74)增高,高密度脂蛋白膽固醇降低(P0.01或P0.05);正常妊娠組,輕、重度子癇前期組血清及胎盤(pán)中oxLDL、LXRα、SREBP-1c的mRNA和(或)蛋白的表達(dá)逐步升高(P0.01或P0.05)。相關(guān)分析顯示,血清LXRα蛋白與血清SREBP-1c蛋白、總膽固醇、三酰甘油、動(dòng)脈硬化指數(shù)呈正相關(guān),與高密度脂蛋白膽固醇呈負(fù)相關(guān)。結(jié)論子癇前期患者血清及胎盤(pán)LXRα表達(dá)明顯增強(qiáng),促進(jìn)其靶基因SREBP-1c的轉(zhuǎn)錄和翻譯,介導(dǎo)母體及胎盤(pán)的脂質(zhì)代謝紊亂。
[Abstract]:Objective to study the liver X receptor alpha in serum and placenta in preeclampsia (LXR alpha) and its target gene of sterol regulatory element binding protein 1c (SREBP-1c) and the relationship between the expression of lipid metabolism and mechanism. 32 cases of patients with preeclampsia were selected method (15 cases of mild preeclampsia and severe preeclampsia, 30 cases and 17 cases) normal non pregnant 30 cases of normal pregnancy. The determination of the level of serum cholesterol, arteriosclerosis index calculation [atherosclerosis index = (total cholesterol and high density lipoprotein cholesterol) / high density lipoprotein cholesterol]; enzyme linked immunosorbent assay (ELISA) determination of the serum protein level of LXR alpha and SREBP-1c; reverse transcriptase polymerase chain the reaction (RT-PCR) and immunohistochemistry (IHC) assay in placenta of oxidized low density lipoprotein (oxLDL), LXR alpha and SREBP-1c mRNA and (or) protein expression level in preeclampsia group. Results compared with the normal pregnancy group, the total cholesterol ([6.78 + 1.56) than (5.20 + 0.96) mmol/L], three glycerol [(3.92 + 1.35) than mmol/L] (2.48 + 0.78), very low density lipoprotein cholesterol [(1.85 + 0.53) than mmol/L] (1.12 + 0.35), low density lipoprotein cholesterol [(4.31 + 1.23) than (3.01 + 0.55 mmol/L]), arterial stiffness index (3.10 + 1.22 to 1.88 + 0.74) increased high density lipoprotein cholesterol (P0.01 or P0.05); normal pregnancy group, severe preeclampsia group, oxLDL in serum and placenta of LXR alpha, SREBP-1c mRNA and (or) protein expression gradually increased (P0.01 or P0.05). Correlation analysis showed that serum LXR protein and serum SREBP-1c protein, total cholesterol, triglyceride was three, atherosclerosis index correlated negatively correlated with high density lipoprotein cholesterol in patients with preeclampsia. Conclusion serum and placental LXR expression increased significantly, promote its target gene SREBP-1c transcription and translation, lipid metabolism mediated by maternal and placental Disorder.
