【摘要】：目的： 通過檢測(cè)YB-1在子宮內(nèi)膜異位癥組織、腹腔液、血清中的表達(dá)情況,探討YB-1在子宮內(nèi)膜異位癥中的發(fā)病機(jī)制,探討YB-1是否可能成為內(nèi)異癥患者一個(gè)潛在的診斷指標(biāo)、治療靶點(diǎn)及預(yù)后標(biāo)志物。 方法： 應(yīng)用免疫組化(IHC)SP法檢測(cè)經(jīng)病理證實(shí)的40例子宮內(nèi)膜異位癥患者中異位內(nèi)膜、在位內(nèi)膜及對(duì)照組30例正常內(nèi)膜中YB-1的表達(dá)情況。采用ELISA檢測(cè)YB-1蛋白在40例子宮內(nèi)膜異位癥患者及30例對(duì)照組患者血清和腹腔液中的表達(dá)變化。探討YB-1與子宮內(nèi)膜異位癥的關(guān)系,并用SPSS17.0軟件對(duì)其結(jié)果進(jìn)行分析,p0.05具有統(tǒng)計(jì)學(xué)意義。 結(jié)果： (1)YB-1蛋白在正常子宮內(nèi)膜、在位子宮內(nèi)膜、異位子宮內(nèi)膜中的表達(dá)呈逐漸增高趨勢(shì)(P0.05),差異有顯著性,主要表達(dá)于子宮內(nèi)膜腺上皮細(xì)胞。間質(zhì)細(xì)胞中也有少量表達(dá)； (2)內(nèi)異癥組YB-1的水平,無論在血清中還是腹腔液中,均高于對(duì)照組(P0.05),且腹腔液中的水平高于血清(P0.05)； (3)在組織中,YB-1的表達(dá)在在位內(nèi)膜中分泌期增生期(P0.05),在異位組和正常內(nèi)膜組無差異(P0.05)；而在血清和腹腔液中,無論內(nèi)異癥組還是正常組,YB-1在分泌期均高于增生期(P0.05),差異有統(tǒng)計(jì)學(xué)意義； (4)YB-1在不同R-AFS分期內(nèi)異癥的子宮內(nèi)膜組織、血清、腹腔液中的表達(dá)水平均不同,Ⅲ-Ⅳ期高于Ⅰ-Ⅱ期,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論： YB-1在子宮內(nèi)膜異位癥組織、血清及腹腔液中高表達(dá),提示YB-1可能參與內(nèi)異癥的發(fā)病,并與內(nèi)異癥病變嚴(yán)重程度相關(guān)。
[Abstract]:Objective: To investigate the pathogenesis of YB-1 in endometriosis by detecting the expression of YB-1 in endometriosis tissue, peritoneal fluid and serum. To explore the possibility of YB-1 as a potential diagnostic marker, therapeutic target and prognostic marker in patients with endometriosis. Methods: The ectopic endometrium in 40 cases of endometriosis confirmed by pathology was detected by immunohistochemical staining (IHC-SP). The expression of YB-1 in eutopic endometrium and 30 normal endometrium was detected by ELISA in serum and peritoneal fluid of 40 patients with endometriosis and 30 patients with normal endometriosis. To investigate the relationship between YB-1 and endometriosis. The results were analyzed by SPSS17.0 software. Results: The expression of YB-1 protein in normal endometrium, eutopic endometrium and ectopic endometrium increased gradually (P 0.05). Mainly expressed in endometrial glandular epithelial cells. (2) the level of YB-1 in the patients with endometriosis was higher than that in the control group (P 0.05), and the level in the peritoneal fluid was higher than that in the control group (P 0.05). (3) the expression of YB-1 in the eutopic endometrium was in the secretory phase of proliferative phase (P0.05), but there was no difference between the ectopic group and the normal group (P0.05). In both serum and peritoneal fluid, the level of YB-1 in the secretory phase was higher than that in the proliferative phase (P 0.05), both in the endometriosis group and the normal group, and the difference was statistically significant.
