【摘要】：目的:上皮細(xì)胞卵巢癌是目前最常見(jiàn)的婦科惡性腫瘤之一,其發(fā)病率每年都在升高。本次研究識(shí)別的重要基因能夠預(yù)測(cè)上皮細(xì)胞卵巢癌患者的預(yù)后,闡釋上皮細(xì)胞卵巢癌的發(fā)病機(jī)制。方法:本研究利用4對(duì)上皮細(xì)胞卵巢癌及其癌旁正常組織的轉(zhuǎn)錄組測(cè)序(RNA-Seq)數(shù)據(jù)進(jìn)行差異基因的篩選,并應(yīng)用Gene Ontology(GO)進(jìn)行富集分析,在TCGA(The Cancer Genome Atlas)在線數(shù)據(jù)庫(kù)中進(jìn)行預(yù)后評(píng)估。結(jié)果:識(shí)別865個(gè)差異表達(dá)基因,通過(guò)功能注釋識(shí)別13個(gè)有意義的GO類型,包括轉(zhuǎn)錄調(diào)控(regulation of transcription)、細(xì)胞黏附(cell adhesion)和生物黏附(biological adhesion)。結(jié)合卵巢癌TCGA在線數(shù)據(jù)庫(kù),發(fā)現(xiàn)2個(gè)重要基因(FBXO 5和CTNNB 1)在高表達(dá)時(shí),不利于上皮細(xì)胞卵巢癌患者的生存;并且在獨(dú)立的另外20對(duì)上皮細(xì)胞卵巢癌及其癌旁正常組織的樣本中,采用PCR證實(shí)了這2個(gè)基因的差異表達(dá)情況。結(jié)論:本研究識(shí)別出的重要基因有助于更好地了解上皮細(xì)胞卵巢癌的發(fā)生和發(fā)展機(jī)制,可能成為預(yù)后判斷的生物標(biāo)志物。
[Abstract]:Objective: epithelial ovarian cancer is one of the most common gynecologic malignancies and its incidence is increasing every year. The important genes identified in this study can predict the prognosis of patients with epithelial ovarian cancer. Methods: in this study, four differentially expressed genes were screened from transcriptome sequencing data of epithelial ovarian carcinoma and its adjacent normal tissues. The enrichment analysis was carried out by using Gene Ontology GO. The prognosis was evaluated in the TCGA(The Cancer Genome Atlas online database. Results: 865 differentially expressed genes were identified. Functional annotations were used to identify 13 meaningful go types, including transcriptional regulation of transcription. Cell adhesion and biological adhesion. Combined with ovarian cancer TCGA online database. It was found that the high expression of FBXO _ 5 and CTNNB _ 1 were harmful to the survival of patients with epithelial ovarian cancer. And in a separate sample of another 20 pairs of epithelial ovarian cancer and its adjacent normal tissue. The differential expression of these two genes was confirmed by PCR. Conclusion: the important genes identified in this study are helpful to better understand the pathogenesis of epithelial ovarian cancer. May be a biomarker for prognostic judgment.
