【摘要】：目的:探討IL-12B基因啟動子區(qū)多態(tài)性與山東濰坊地區(qū)漢族育齡期婦女原因不明復(fù)發(fā)性流產(chǎn)(URSA)的相關(guān)性。方法:采用直接測序法檢測83例原因不明復(fù)發(fā)性流產(chǎn)孕婦和90例正常孕婦IL-12B基因的啟動子區(qū)的多態(tài)位點,并分析IL-12B基因多態(tài)性與原因不明復(fù)發(fā)性流產(chǎn)的關(guān)系。結(jié)果:IL-12B的啟動子區(qū)檢測到2個單核苷酸多態(tài)性(SNPs)(-671G/T和-405G/T)。2個SNPs緊密連鎖,多態(tài)性分布符合Hardy-Weinberg平衡。URSA組和對照組的基因型和等位基因頻率分布比較,差異無統(tǒng)計學(xué)意義(P0.05)。單體型分析顯示,-671T/-405G頻率在URSA組和正常對照組分別為0.65和0.41,URSA組顯著高于正常對照組,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:正常孕婦IL-12B基因啟動子區(qū)存在多態(tài)性變異,其中-671G/T和-405G/T構(gòu)成的單體型-671T/-405G可能與URSA的易感性有關(guān)。
[Abstract]:Objective: to investigate the polymorphism of IL-12B gene promoter region and Ursa of unexplained recurrent abortion among Han women of childbearing age in Weifang area of Shandong Province. Methods: the polymorphic loci of IL-12B gene promoter in 83 pregnant women with unexplained recurrent abortion and 90 normal pregnant women were detected by direct sequencing. The relationship between IL-12B gene polymorphism and unexplained recurrent abortion was analyzed. Results two single nucleotide polymorphisms (SNPs) were detected in the promoter region of 1: IL-12B. -671G / T and -405G / T. 2 SNPs closely linked. The polymorphism distribution was consistent with the comparison of genotype and allele frequency distribution between the Hardy-Weinberg equilibrium. Ursa group and the control group. Haplotype analysis showed that the frequencies of -671T / -405G in URSA group and normal control group were 0.65 and 0.41, respectively. The URSA group was significantly higher than the normal control group, the difference was statistically significant (P 0.05). Conclusion: there is polymorphism in the promoter region of IL-12B gene in normal pregnant women. The haplotype -671T / -405G composed of -671G / T and -405G / T may be related to the susceptibility of URSA.
