苯磺酸氨氯地平抑制高糖誘導(dǎo)的H9C2細(xì)胞凋亡研究

【摘要】:目的利用細(xì)胞實(shí)驗(yàn)探討高糖培養(yǎng)對(duì)H9C2細(xì)胞凋亡的影響,并探討苯磺酸氨氯地平的保護(hù)作用。方法體外培養(yǎng)大鼠心肌細(xì)胞H9C2,分為5mmol/L糖培養(yǎng)組(G1)、25mmol/L糖培養(yǎng)組(G2)、50mmol/L糖培養(yǎng)組(G3)和25mmol/L糖培養(yǎng)組加鈣離子通道抑制劑絡(luò)活喜保護(hù)組(G2+N)、50mmol/L糖培養(yǎng)組加絡(luò)活喜保護(hù)組(G3+N)5組,每組分設(shè)48h(a)、72h(b)培養(yǎng)兩個(gè)亞組共10組。AnnexinV/PI結(jié)合流式細(xì)胞儀檢測(cè)細(xì)胞凋亡率,熒光染色觀察[Ca2+]i。結(jié)果細(xì)胞凋亡率隨著高糖刺激時(shí)間和高糖濃度的增加而逐漸增高,加絡(luò)活喜組細(xì)胞的凋亡率顯著降低(P0.05)。G2、G3組單細(xì)胞平均[Ca2+]i活性測(cè)定熒光值均較G1組升高(P0.05);G2+N、G3+N組單細(xì)胞平均[Ca2+]i活性測(cè)定熒光值分別較G2、G3組降低(P0.05)。各a、b亞組間單細(xì)胞平均[Ca2+]i活性測(cè)定熒光值差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論高糖培養(yǎng)H9C2細(xì)胞可增加[Ca2+]i從而導(dǎo)致細(xì)胞凋亡,苯磺酸氨氯地平可抑制Ca2+內(nèi)流從而抑制細(xì)胞凋亡。
[Abstract]:Objective to investigate the effect of high glucose on the apoptosis of H9C2 cells and to explore the protective effect of amlodipine sulfonate. The myocardial cells of H9C2 rats were cultured in vitro, divided into 5mmol/L culture group (G1), sugar sugar 25mmol/L culture group (G2), 50mmol/L group (G3) and glucose 25mmol/L glucose group plus calcium channel inhibitor (G2+N), amlodipine group 50mmol/L group and amlodipine sugar culture protection group (G3+N) 5 each group is divided into group 48h (a), 72h (b) training of two sub group 10 group. The apoptosis rate was detected by AnnexinV/PI combined with flow cytometry, and [Ca2+]i was observed by fluorescence staining. Results the apoptosis rate increased with high glucose time and high sugar concentration gradually increased, apoptosis rate of cells with amlodipine group decreased significantly (P0.05). The average [Ca2+]i activity of single cells in group G2 and group G3 was higher than that in group G1 (P0.05), and the mean value of [Ca2+]i activity in G2+N group and G3+N group was lower than those in G2 and G3 group (P0.05). There was no significant difference in the fluorescence value of the average [Ca2+]i activity of single cells between the A and B subgroups (P0.05). Conclusion high sugar H9C2 cells can increase [Ca2+]i and lead to cell apoptosis. Amlodipine besylate inhibits Ca2+ influx and inhibits apoptosis.
