促紅細(xì)胞生成素與黃芪對新生兒窒息后血清誘導(dǎo)人腎小管細(xì)胞TLR 4 /MyD88表達(dá)的影響

目的:研究促紅細(xì)胞生成素與黃芪對新生兒窒息后血清誘導(dǎo)人腎小管細(xì)胞TLR4/MyD88 (Toll-like receptors 4/myeloid differentiation factor 88)表達(dá)的影響方法:(1)以HK-2細(xì)胞(Human renal proximal tubular cell,人近曲腎小管上皮細(xì)胞)作為研究對象。(2)以足月重度窒息(生后Apgar評分均小于4分)新生兒第一個(gè)24小時(shí)血清(20%體積分?jǐn)?shù))作為致病因素。(3)實(shí)驗(yàn)分四組,即對照組、窒息組、EPO(erythropoietin,促紅細(xì)胞生成素)組、黃芪(Astragalus, AS)組。(4)檢測指標(biāo):倒置相差顯微鏡下觀察HK-2細(xì)胞形態(tài)變化;流式細(xì)胞儀(Flow Cytometry, FCM)檢測各組細(xì)胞TLR4特異性熒光抗體陽性結(jié)合率表達(dá)情況;采用免疫組織化學(xué)法(SP法)檢測各組細(xì)胞MyD88表達(dá)情況。(5)統(tǒng)計(jì)學(xué)分析:數(shù)據(jù)均用x±s表示,采用one-way ANOVA分析,組間比較用LSD法,運(yùn)用SPSS14.0軟件進(jìn)行統(tǒng)計(jì)學(xué)處理,P<0.05為差異具有統(tǒng)計(jì)學(xué)意義。結(jié)果:(1)倒置相...
