SARM抑制TRIF在RSV感染后氣道炎癥及氣道高反應性作用機制研究

第一部分RSV感染后SARM及TRIF表達水平研究目的:RSV是引起5歲以下兒童下呼吸道感染最常見的病毒病原體。Toll樣受體(TLR)信號通路與RSV致病機制密切相關。RSV感染后可通過激活TLR-TRIF依賴的信號通路引起氣道炎癥及氣道高反應性。SARM為TRIF的負性調節(jié)因子,對RSV感染所致的氣道炎癥及氣道高反應性(AHR)影響作用尚不清楚。本實驗目的是研究RSV感染后SARM及TRIF的表達情況。方法:體外培養(yǎng)人類氣道上皮細胞株9HTEo,RSV以MOI=10感染9HTEo細胞,分別在12h、24h、36h、48h和72h后收集細胞,提取全蛋白,Western blot方法檢測感染后不同時間點SARM及TRIF的水平變化。RSV或紫外線滅活的RSV (UV RSV)感染6~8周齡雌性BALB/c小鼠感染,分別在感染的第3、5、7天處死,取小鼠肺組織,提取肺組織總蛋白,Western blot方法檢測SARM及TRIF的變化情況。利用RES進行進一步干預,將6~8周齡雌性BALB/c小鼠分為對照組、RSV+PBS組、RSV+RES組,每組各5只,感染后第5天Western bl...
