【摘要】：目的 探討T細(xì)胞受體保守域α鏈基因(TCRCα)-560C/T,575A/G位點(diǎn)多態(tài)性與過(guò)敏性紫癜(HSP)發(fā)病之間的關(guān)系。 方法 應(yīng)用聚合酶鏈反應(yīng)-限制內(nèi)切酶片段長(zhǎng)度多態(tài)分析(PCR-PFLP)方法結(jié)合瓊脂糖凝膠電泳技術(shù),對(duì)36例過(guò)敏性紫癜患兒和60例正常對(duì)照組兒童進(jìn)行TCRCα-560C/T,575A/G位點(diǎn)基因多態(tài)性分析。 結(jié)果 (1) TCRCα基因-560位點(diǎn)的CC、CT、TT三種基因型頻率在紫癜組和正常組中比較無(wú)顯著性差異(x2=5.39,P0.05),而該位點(diǎn)的等位基因頻率C和T在紫癜組和正常組之間差異有顯著意義(x2=4.69,P0.05)。 (2)TCRCα基因-575位點(diǎn)的AA、AG、GG三種基因型頻率在紫癜組和正常組中比較無(wú)顯著性差異(x2=1.07,P0.05),該位點(diǎn)的等位基因頻率A和G在紫癜組和正常組差異無(wú)顯著性(x2=1.01,P0.05)。 結(jié)論 (1) TCRCα-560C/T基因多態(tài)性變化與HSP的發(fā)生可能存在一定關(guān)聯(lián)。 (2)TCRCα-575A/G基因多態(tài)性變化與HSP的發(fā)生無(wú)明顯關(guān)聯(lián)性。
[Abstract]:Aim to investigate the relationship between T cell receptor conserved domain 偽 chain gene (TCRC 偽) 560C / T, 575A 偽 G polymorphism and the pathogenesis of (HSP) in Henoch-Schonlein purpura (Henoch-Schonlein purpura). Methods 36 children with Henoch-Schonlein purpura (Henoch-Schonlein purpura) and 60 normal children were studied with polymerase chain reaction-restriction endonuclease fragment length polymorphism (PCR-PFLP) combined with agarose gel electrophoresis. Polymorphism analysis of 575A/G locus gene. Results (1) there was no significant difference in the frequencies of three genotypes of CC,CT,TT at site-560of TCRC 偽 gene between purpura group and normal group (x2, 5.39, P0.05). The allele frequencies C and T of this locus were significantly different between purpura group and normal group (x2 / 4.69, P0.05). (2) there was no significant difference in the frequencies of AA,AG,GG genotypes at site-575 of TCRC 偽 gene between purpura group and normal group (x2? 1.07, P0.05). There was no significant difference in allele frequencies A and G between Henoch-Schonlein purpura group and normal group (x2? 1.01, P0.05). Conclusion (1) the polymorphism of TCRC 偽-560C/T gene may be associated with the occurrence of HSP. (2) there was no significant correlation between the polymorphism of TCRC 偽-575A/G gene and the occurrence of HSP.
