RANKL、OPG在小兒骨瘤樣病變(單純性骨囊腫、動(dòng)脈瘤樣骨囊腫)組織中的表達(dá)研究

【摘要】:【目的】通過對(duì)小兒骨瘤樣病變(單純性骨囊腫、動(dòng)脈瘤樣骨囊腫)組織中OPG、RANKL蛋白表達(dá)水平的檢測(cè),并與正常的骨組織作對(duì)照,分析這兩種蛋白在不同溶骨性病變中的表達(dá)是否存在差異性及意義。 【方法】共收集動(dòng)脈瘤樣骨囊腫標(biāo)本23例、單純性骨囊腫標(biāo)本31例,正常骨組織標(biāo)本14例。采用免疫組化SABC方法研究單純性骨囊腫、動(dòng)脈瘤樣骨囊腫中OPG、 RANKL的表達(dá)并分析其差異性。應(yīng)用Imagepro-Plus6.0圖像處理軟件及數(shù)據(jù)處理軟件SPSS17.0對(duì)圖像及所得數(shù)據(jù)經(jīng)行分析。 【結(jié)果】在單純性骨囊腫及動(dòng)脈瘤樣骨囊腫中,RANKL蛋白的表達(dá)量明顯高于正常骨組織的表達(dá),具有統(tǒng)計(jì)學(xué)意義,其中動(dòng)脈瘤樣骨囊腫RANKL蛋白的表達(dá)量較單純性骨囊腫增高,有統(tǒng)計(jì)學(xué)意義;兩種瘤樣病變中OPG蛋白的表達(dá)量低于正常骨組織的表達(dá),有統(tǒng)計(jì)學(xué)意義,其中動(dòng)脈瘤樣骨囊腫OPG稍高于單純性骨囊腫,無(wú)統(tǒng)計(jì)學(xué)意義。RANKL/OPG比值在動(dòng)脈瘤樣骨囊腫>單純性骨囊腫㧐正常組織,有統(tǒng)計(jì)學(xué)意義。 【結(jié)論】RANKL、OPG平衡的嚴(yán)重破壞是導(dǎo)致機(jī)體發(fā)生自發(fā)性骨質(zhì)破壞的原因;RANKL/OPG比值的輕微升高不會(huì)導(dǎo)致機(jī)體發(fā)生溶骨性破壞,推測(cè)RANKL/OPG比值達(dá)到一定界點(diǎn)后才會(huì)導(dǎo)致機(jī)體發(fā)生骨破壞,,此比值界點(diǎn)有待進(jìn)一步研究。初步表明RANKL的高表達(dá)與動(dòng)脈瘤樣骨囊腫的復(fù)發(fā)密切相關(guān)。
[Abstract]:[objective] to detect the expression of OPG,RANKL protein in osteomatous lesions (simple bone cysts, aneurysm-like bone cysts) of children, and to compare it with normal bone tissues. To analyze the difference and significance of the expression of these two proteins in different osteolytic lesions. [methods] 23 cases of aneurysm like bone cyst, 31 cases of simple bone cyst and 14 cases of normal bone tissue were collected. Immunohistochemical SABC method was used to study the expression of OPG, RANKL in simple bone cysts and aneurysm like bone cysts. Imagepro-Plus6.0 image processing software and data processing software SPSS17.0 are used to analyze the image and the data. [results] the expression of RANKL protein in simple bone cysts and aneurysm like bone cysts was significantly higher than that in normal bone tissues, and the expression of RANKL protein in aneurysm like bone cysts was higher than that in simple bone cysts. Statistical significance; The expression of OPG protein in two kinds of tumor-like lesions was lower than that in normal bone tissue, and the expression of OPG in aneurysm-like bone cysts was slightly higher than that in simple bone cysts. There was no statistical significance. The ratio of RANKL/OPG in aneurysm like bone cyst was higher than that in simple bone cyst. Normal tissue, with statistical significance. [conclusion] the serious destruction of RANKL,OPG balance is the cause of spontaneous bone destruction. The slight increase of RANKL/OPG ratio will not lead to osteolytic destruction. It is speculated that bone destruction will occur only when the RANKL/OPG ratio reaches a certain point, which should be further studied. The high expression of RANKL is closely related to the recurrence of aneurysm like bone cyst.
