生命早期食物過敏對幼鼠內(nèi)臟痛敏感性影響及結(jié)腸P物質(zhì)異常表達(dá)在其形成中作用
[Abstract]:Objective to establish the (FA) model of food allergy to ovalbumin (OVA) in young SD rats and to investigate the effect of FA on visceral pain sensitivity and the abnormal expression of substance P (SP) in colon during the formation of visceral hyperalgesia Gao Min in FA young rats. Methods Twenty 3-week-old female SD rats were randomly divided into FA group (n = 10) and control group (NS group, n = 10). SD young rats were sensitized by low dose OVA intraperitoneal injection and stimulated by high dose OVA. OVA-FA; was used to detect OVA IgE content in serum of SD young rats. The pathological changes of intestinal mucosa and the infiltration of eosinophils and mast cells were observed to evaluate the FA model of young rats. The visceral pain sensitivity was evaluated by observing the (EOMA) discharges of the oblique ventral muscles of the two groups after (CRD) stimulation under different pressures. Immunohistochemical staining and computer image analysis system were used for semi-quantitative analysis of integrated optical density (OSD) of colon SP. SPSS13.0 software package was used for statistical analysis, 偽 -0. 05 was the significant test standard. Results the young rats in FA group had different degrees of hair erecting, hair losing luster, decreased activity or prone immobility, scratching of ear, restlessness or restlessness and defecation, compared with control group, the content of serum OVA-IgE in FA group was higher than that in control group. The number of eosinophilic granulocytes and mast cells and the percentage of degranulation of mast cells in jejunal mucosa were significantly increased (P < 0.01). When the CRD was 0 ~ 15 ~ 30 ~ 45 ~ 60 ~ 75mmHg, the (uV) of the external oblique muscle in the FA group was 17.74 鹵0.72 ~ 18.63 鹵1.72t 22.55 鹵1.7028.63 鹵7.34 鹵37.97 鹵7.34 ~ 37.26 鹵8.4mm Hg, respectively, which was 17.43 鹵1.1817.27 鹵1.167.73 鹵1.42n 19.55 鹵3.523.29 鹵5.46n 25.20 鹵4.75 respectively. With the increase of CRD, the discharge amplitude of the oblique ventral muscle increased significantly (P < 0. 01). The SP optical density of colon in the FA group was 247.12 鹵90.83103.90 鹵58.94, respectively (P < 0. 01), and there was a significant difference between the two groups (P < 0. 01). Conclusion the FA model of young SD rats can be established by low dose intraperitoneal injection of OVA and high dose of OVA by intraperitoneal injection. Early FA could cause the amplitude of ventral obliquus muscle discharge and the expression of SP in colon increased, and the chronic visceral pain was hypersensitive. The high sensitivity of visceral pain induced by FA in early life may be related to the increased expression of mast cells in intestinal mucosa, degranulation and abnormal expression of intestinal SP.
