依達拉奉對高膽紅素血癥新生大鼠血清NSE的影響

【摘要】:目的:探討依達拉奉干預對高膽紅素血癥新生大鼠血清神經元特異性烯醇化酶(neuron-specific enolase,NSE)水平的影響。方法:將72只7日齡SD大鼠隨機地分到H、E、C組,H組為高膽紅素血癥組(n=24)、E組為依達拉奉干預組(n=24)、C組為正常對照組(n=24)。每組又根據處死時間的不同隨機分為3個亞組:12h組、24h組、48h組,每亞組8只。采用腹腔注射的方法,給H組、E組新生大鼠按100μg/g的劑量注入膽紅素1次建立高膽紅素血癥動物模型,用同樣的方法給C組新生大鼠按100μg/g的劑量注入生理鹽水1次,造模后立即給予E組依達拉奉干預,劑量為2mg/kg/次,一日一次。觀察各組新生大鼠有無行為異常如有無抽搐、活動減少、對外界刺激的逃避反應減弱等,并于各時間點取血及腦組織。蘇木素-伊紅染色(hematoxylin-eosin staining,HE)觀察各組新生大鼠腦組織的病理改變,用酶聯免疫法(ELISA法)測定血清中NSE。結果:1.實驗前,H、E、C3組新生大鼠行為均無明顯的異常;造模后H組、E組新生SD大鼠出現行為異常,如抽搐、活動減少、對外界刺激的逃避反應減弱等,但E組在行為異常方面較高膽紅素血癥組輕;C組新生SD大鼠在整個實驗中均未出現行為方面的明顯異常。2.HE染色結果:C組海馬區(qū)細胞排列整齊,細胞形態(tài)結構完整,無明顯細胞水腫;H組海馬區(qū)細胞排列略紊亂,細胞水腫明顯,有明顯炎性細胞浸潤;E組腦組織可見少許神經細胞水腫,但在各時間點上腦組織病理改變均較H組輕。3.在各時間點上,H組新生大鼠血清NSE水平較C組高,P0.05,差異具有統計學意義;在各時間點上,E組新生大鼠血清NSE水平較H組低,但仍高于C組,P0.05,差異有統計學意義。結論:新生大鼠高膽紅素血癥時,血清NSE水平增加,給予依達拉奉干預,可以降低血清NSE水平,表明依達拉奉對高膽紅素血癥腦損傷新生大鼠有神經保護作用。
[Abstract]:Aim: to investigate the effect of Edaravone on serum neuron specific enolase neuron-specific enolase (NSE) levels in neonatal rats with hyperbilirubinemia. Methods: Seventy-two 7-day-old SD rats were randomly divided into two groups: group H was hyperbilirubinemia group (n = 24), group E was Edaravone intervention group (n = 24), group C was normal control group (n = 24). According to the time of death, each group was randomly divided into 3 subgroups: 12 h group, 24 h group, 48 h group, 8 rats in each subgroup. The hyperbilirubinemia animal model was established by intraperitoneal injection of bilirubin at 100 渭 g / g dose to the neonatal rats in group H, and normal saline was injected to the newborn rats in group C at a dose of 100 渭 g / g. Group E was treated with Edaravone at the dose of 2mg/kg/ once a day. We observed whether the newborn rats in each group had behavioral abnormalities, such as convulsions, decreased activity, decreased escape response to external stimulation, and took blood and brain tissue at each time point. Hematoxylin-eosin staining (hematoxylin-eosin) was used to observe the pathological changes in the brain tissue of neonatal rats in each group, and the serum NSE was determined by enzyme-linked immunosorbent assay (Elisa). The result is 1: 1. Before the experiment, there was no obvious abnormality in the behavior of newborn rats in group C 3, and abnormal behavior of newborn SD rats in group E of H group after modeling, such as convulsion, decreased activity, weak escape response to external stimuli, etc. However, there was no obvious behavioral abnormality in group E with hyperbilirubinemia. 2. The results of HE staining showed that the hippocampal cells in group E were arranged neatly, and the morphology and structure of the cells were intact. There was no obvious cell edema in the hippocampal area of group H, but there were some neuronal edema in the brain tissue of group E, but the pathological changes of brain tissue in group H were lighter than those in group H at different time points. The serum NSE level in group H was significantly higher than that in group C (P 0.05), and that in group E was lower than that in group H at each time point, but still higher than that in group C (P 0.05). Conclusion: the level of serum NSE in neonatal rats with hyperbilirubinemia is increased. Edaravone intervention can decrease the level of serum NSE, indicating that Edaravone has neuroprotective effect on neonatal rats with hyperbilirubinemia brain injury.
