新疆南部地區(qū)維吾爾族兒童尿路結(jié)石與維生素D受體基因、降鈣素受體基因多態(tài)性相關(guān)性研究

【摘要】:目的:探討維生素D受體(VDR)基因與降鈣素受體(CTR)基因多態(tài)性與新疆南部地區(qū)維吾爾族小兒尿路結(jié)石相關(guān)性,闡明分子遺傳學(xué)規(guī)律。方法:維吾爾族尿路結(jié)石患兒(病例組)74例,維吾爾族正常兒童(對(duì)照組)103例納入此研究,應(yīng)用聚合酶鏈反應(yīng)-限制性片段長(zhǎng)度多態(tài)性分析研究VDR基因FokI和ApaI位點(diǎn)多態(tài)性及CTR基因多態(tài)性與新疆南部地區(qū)維吾爾族小兒尿路結(jié)石相關(guān)性。結(jié)果: VDR基因FokI位點(diǎn)FF、 Ff、 ff三種基因型在病例組及對(duì)照組之間分布有統(tǒng)計(jì)學(xué)意義(χ~2=7.818,P<0.05),,基因型Ff在病例組占58.1%,明顯高于對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。VDR基因ApaI位點(diǎn)AA、Aa、aa三種基因型在病例組中的分布相似于對(duì)照組,分布無(wú)統(tǒng)計(jì)學(xué)差異。CTR基因各種基因型在病例組與對(duì)照組之間的分布有統(tǒng)計(jì)學(xué)意義(P<0.05)。ApaI位點(diǎn)基因型在病例組男女之間分布有差異,但VDR基因FokI位點(diǎn)、CTR基因在性別之間無(wú)基因型分布差異。結(jié)論: VDR基因FokI位點(diǎn)多態(tài)性與CTR基因多態(tài)性將來(lái)可作為維吾爾族小兒泌尿系結(jié)石的候選基因。
[Abstract]:Objective: to investigate the relationship between the polymorphism of vitamin D receptor (VDR) gene and calcitonin receptor (CTR) gene and urinary tract stones in Uygur children in southern Xinjiang, and to elucidate the rules of molecular genetics. Methods: 74 Uygur children with urinary tract stones and 103 normal Uygur children were included in the study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to study the association of VDR gene FokI and ApaI locus polymorphism and CTR gene polymorphism with urinary tract stones in Uygur children in southern Xinjiang. Results: the distribution of three FF, Ff, ff genotypes in FokI locus of VDR gene was statistically significant between the case group and the control group (蠂 ~ (2) 7.818) (P < 0.05). The percentage of genotype F _ f in the case group was 58.1%, which was significantly higher than that in the control group. There was significant difference in the distribution of the three genotypes of ApaI locus of P < 0.05).VDR gene in the case group, and the distribution of the three genotypes in the case group was similar to that in the control group. There was no statistical difference in the distribution of all genotypes of CTR gene between the case group and the control group. There was a significant difference in the distribution of the genotype P < 0.05).ApaI locus between the male and the female in the case group. However, there was no genotypic distribution difference between genders at FokI locus of VDR gene. Conclusion: VDR gene FokI locus polymorphism and CTR gene polymorphism can be used as candidate genes for Uygur children urinary calculi in the future.
