【摘要】：目的探討嬰幼兒毛細支氣管炎中miRNA-26b、抵抗素的變化。方法收集毛細支氣管炎患兒的血液,以正常兒童為對照,通過酶聯(lián)免疫吸附(ELISA)法測定血清中白介素(IL)-8、抵抗素濃度,并利用實時定量PCR技術(shù)測定血液淋巴細胞中miRNA-26b、抵抗素mRNA表達,利用miRNA-26b mimics干預人肺上皮細胞A549,通過實時定量PCR技術(shù)測定miRNA-26b、抵抗素mRNA及細胞上清液中IL-8蛋白濃度變化。結(jié)果毛細支氣管炎患兒外周血清中IL-8、抵抗素濃度均升高,血液淋巴細胞中miRNA-26b、抵抗素mRNA表達增加;經(jīng)過干預的A549細胞miRNA-26b、抵抗素mRNA及細胞上清液中IL-8蛋白濃度均有增加。結(jié)論 miRNA-26b、抵抗素在毛細支氣管炎患兒血液淋巴細胞中存在差異表達,而這可能參與了炎癥反應的發(fā)生。
[Abstract]:Objective to investigate the changes of miRNA-26 b and resistin in infants with bronchiolitis. Methods Blood samples from children with bronchiolitis were collected and compared with normal children. The concentrations of interleukin-8 and resistin in serum were determined by Elisa, and the expression of miRNA-26b and resistin mRNA in blood lymphocytes was measured by real-time quantitative PCR technique. Human lung epithelial cells A549 were treated with miRNA-26b mimics. The concentration of miRNA-26b, resistin mRNA and IL-8 protein in supernatant of human lung epithelial cells were measured by real-time quantitative PCR. Results the concentrations of IL-8 and resistin in peripheral serum of children with bronchiolitis were all increased, miRNA-26b and resistin mRNA expression in blood lymphocytes were increased, and miRNA-26b, resistin mRNA and IL-8 protein in supernatant of A549 cells were increased after intervention. Conclusion miRNA-26b, resistin is differentially expressed in blood lymphocytes of children with bronchiolitis, which may be involved in inflammatory reaction.
