益生菌對(duì)肥胖大鼠血脂及胰島素相關(guān)指標(biāo)的影響

【摘要】:目的:1.高脂飲食構(gòu)建單純性肥胖大鼠模型;2.觀察空白對(duì)照組、高脂對(duì)照組、嗜酸乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組大鼠肥胖相關(guān)指標(biāo)(體重、腹圍、身長(zhǎng)、Lee’s指數(shù))及胰島素相關(guān)指標(biāo)(空腹血糖、空腹血清胰島素、胰島素敏感指數(shù)、胰島素分泌指數(shù)、胰島素抵抗指數(shù))的變化,探討肥胖、血脂、胰島素相關(guān)指標(biāo)的關(guān)系及益生菌(嗜酸性乳桿菌、短雙歧桿菌)對(duì)肥胖大鼠血脂及胰島素相關(guān)指標(biāo)的干預(yù)作用。 方法:3周齡,體質(zhì)量50±5gSD雄性大鼠50只,隨機(jī)分為2組:空白對(duì)照組(N組)10只給予普通飼料喂養(yǎng),高脂飲食組40只給予高脂飼料喂養(yǎng)。喂養(yǎng)4周,高脂飲食組中以超出空白對(duì)照組平均體重的20%作為試驗(yàn)用肥胖大鼠,高脂飲食組4只未達(dá)標(biāo)準(zhǔn),予以剔除,剩余36只肥胖大鼠隨即分為高脂對(duì)照組(A組)、嗜酸乳桿菌干預(yù)組(B組)、短雙歧桿菌干預(yù)組(C組),每組12只。N組每天給予普通飼料+1ml/100g體重生理鹽水灌胃;A組每天給予高脂飼料+1ml/100g體重生理鹽水灌胃;B組每天給予高脂飼料+1ml/100g體重嗜酸乳桿菌菌液灌胃;C組每天給予高脂飼料+1ml/100g體重短雙歧桿菌菌液灌胃,飼養(yǎng)4周。每周稱量體重1次,根據(jù)體重變化調(diào)整灌胃量。實(shí)驗(yàn)結(jié)束后測(cè)定大鼠體質(zhì)量、腹圍、體長(zhǎng),并計(jì)算Lee’s肥胖指數(shù),檢測(cè)空腹血糖(FPG)、血清膽固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、空腹血清胰島素(Fins),并計(jì)算胰島素敏感指數(shù)(IAI)、胰島素分泌指數(shù)(IS)、胰島素抵抗指數(shù)(IRI)。所得實(shí)驗(yàn)數(shù)據(jù)均采用SPSS17.0統(tǒng)計(jì)軟件進(jìn)行分析。 結(jié)果: 1.肥胖大鼠造模期間與空白對(duì)照組比較,給予高脂飲食組的大鼠體態(tài)明顯增大,,活動(dòng)量減少,飲水量、排尿量增多。給予益生菌灌胃干預(yù)4周后與高脂對(duì)照組比較,體態(tài)增長(zhǎng)較慢,活動(dòng)量略有增多,飲水量、排尿量減少。 2.肥胖相關(guān)指標(biāo)的比較:空白對(duì)照組、高脂對(duì)照組、嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組體重(g)分別為:208.60±20.97、341.60±24.57、324.80±14.78、320.20±20.60;腹圍(cm)分別為:13.84±0.80、17.14±0.56、14.96±0.79、15.00±1.13;身長(zhǎng)(cm)分別為:23.96±0.33、24.16±0.44、24.14±0.56、23.86±0.37;Lee’s指數(shù)(103)分別為:2904.12±310.65、4715.24±360.19、4486.21±208.37、4474.68±301.61。各組大鼠體重、腹圍、Lee’s指數(shù)比較,高脂對(duì)照組明顯高于空白對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組明顯低于高脂對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,但嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組比較無(wú)明顯差異(P0.05),差別無(wú)統(tǒng)計(jì)學(xué)意義;各組大鼠身長(zhǎng)比較無(wú)明顯差異(P0.05),差別無(wú)統(tǒng)計(jì)學(xué)意義。 3.血脂相關(guān)指標(biāo)比較:空白對(duì)照組、高脂對(duì)照組、嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組TC(mg/dl)分別為:48.40±7.87、69.42±6.09、53.21±10.17、56.19±9.22;TG(mg/dl)分別為:8.23±2.11、29.19±5.37、18.81±6.32、15.32±5.19;HDL(mg/dl)分別為:17.40±2.97、11.64±2.03、14.47±2.22、14.40±2.17;LDL(mg/dl)分別為:12.67±1.68、17.23±2.69、14.41±2.13、13.29±1.78。各組大鼠TC、TG、LDL比較,高脂對(duì)照組明顯高于空白對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組明顯低于高脂對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組比較無(wú)明顯差異(P0.05),差別無(wú)統(tǒng)計(jì)學(xué)意義;各組大鼠HDL比較,高脂對(duì)照組明顯低于空白對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組明顯高于高脂對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組比較無(wú)明顯差異(P0.05),差別無(wú)統(tǒng)計(jì)學(xué)意義。 4.胰島素相關(guān)指標(biāo)比較:空白對(duì)照組、高脂對(duì)照組、嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組FPG(mmol/L)分別為:7.89±1.71、12.43±1.27、10.08±0.93、9.40±1.34;Fins(mIU/L)分別為:5.03±0.99、13.28±0.78、8.70±0.87、6.42±1.14;IAI分別為:-1.57±0.17、-2.21±0.03、-1.93±0.06、-1.77±0.12;IS分別為:0.66±0.12、1.08±0.17、0.87±0.12、0.69±0.11;IRI分別為:1.80±0.61、7.31±0.54、3.90±0.52、2.72±0.76。各組大鼠空腹血糖比較,高脂對(duì)照組明顯高于空白對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組明顯低于高脂對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,但嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組比較無(wú)明顯差異(P0.05),差別無(wú)統(tǒng)計(jì)學(xué)意義;血清胰島素、胰島素敏感指數(shù)、胰島素分泌指數(shù)、胰島素抵抗指數(shù)比較,高脂對(duì)照組明顯高于空白對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組明顯低于高脂對(duì)照組(P0.05),差別有統(tǒng)計(jì)學(xué)意義,嗜酸性乳桿菌干預(yù)組、短雙歧桿菌干預(yù)組比較,短雙歧桿菌干預(yù)組明顯低于嗜酸性乳桿菌干預(yù)組(P0.05),差別有統(tǒng)計(jì)學(xué)意義。 結(jié)論: 1.給予高脂飲食喂養(yǎng)4周后成功構(gòu)建肥胖大鼠模型。 2.肥胖大鼠發(fā)生了血脂紊亂、空腹血糖升高、高胰島素血癥及胰島素抵抗。 3.嗜酸性乳桿菌與短雙歧桿菌均具有降低肥胖大鼠血脂、空腹血糖,改善胰島素抵抗相關(guān)指標(biāo)作用,且不影響大鼠的正常生長(zhǎng)發(fā)育。 4.在減輕體重、改善血脂異常方面嗜酸性乳桿菌與短雙歧桿菌菌種療效相當(dāng),在改善胰島素抵抗相關(guān)指標(biāo)方面,短雙歧桿菌優(yōu)于嗜酸性乳桿菌。

Methods: 3 weeks of age, 50 male rats with body mass of 50 5gSD were randomly divided into 2 groups: 10 rats in the blank control group (group N) were fed with ordinary feed, and 40 of the high fat diet group were fed with high fat feed.
