早產(chǎn)兒臍血中γδ T淋巴細(xì)胞功能變化特征的研究

【摘要】:目的檢測分析早產(chǎn)兒和足月兒臍帶血中γδT細(xì)胞的數(shù)量、比例及相關(guān)功能變化特征,探討早產(chǎn)兒人群的γδT細(xì)胞功能變化對其抵抗病毒感染的意義。方法采用流式細(xì)胞術(shù)檢測臍帶血單個(gè)核細(xì)胞(cord blood mononuclear cell,CBMC)中γδT細(xì)胞的數(shù)量比例;流感病毒感染刺激早產(chǎn)兒和足月兒CBMC 24 h,流式細(xì)胞術(shù)測定γδT細(xì)胞中細(xì)胞因子Perforin、IFN-γ和TNF-α的表達(dá)情況;采用異戊烯焦磷酸(isopentenyl pyrophosphate,IPP)聯(lián)合IL-2刺激擴(kuò)增CBMC中γδT細(xì)胞,比較早產(chǎn)兒和足月兒γδT細(xì)胞的擴(kuò)增效率。結(jié)果早產(chǎn)兒γδT細(xì)胞在CBMC中的數(shù)量及比例較足月兒明顯減少;流感病毒刺激后,早產(chǎn)兒和足月兒γδT細(xì)胞的Perforin、IFN-γ和TNF-α的表達(dá)都有顯著升高,但早產(chǎn)兒倍增幅度明顯不及足月兒;經(jīng)IPP和IL-2聯(lián)合刺激后,臍帶血中γδT細(xì)胞都有所增殖,但早產(chǎn)兒的擴(kuò)增效率明顯低于足月兒。結(jié)論早產(chǎn)兒γδT細(xì)胞存在數(shù)量及功能的不足,這可能是造成早產(chǎn)兒更易患感染性疾病的重要因素之一。
[Abstract]:Objective to detect and analyze the changes of 緯 未 T cells in cord blood of premature infants and term infants. To investigate the significance of 緯 未 T cell function changes in premature infants against virus infection. Methods the percentage of 緯 未 T cells in cord blood mononuclear cell (blood mononuclear cell blood) was detected by flow cytometry. Influenza virus infection stimulated CBMC of premature and term infants for 24 h, the expression of cytokines Perforinine IFN- 緯 and TNF- 偽 in 緯 未 T cells was detected by flow cytometry, and 緯 未 T cells in CBMC were amplified by isopentenyl isopentenyl phosphate IPP combined with IL-2 stimulation, and the expression of IFN- 緯 and TNF- 偽 in 緯 未 T cells were detected by flow cytometry. Results the number and proportion of 緯 未 T cells in CBMC of premature infants were significantly lower than those of full-term infants, and the expression of Perforintin-IFN- 緯 and TNF- 偽 in preterm and full-term infants were significantly increased after influenza virus stimulation. After IPP and IL-2 stimulation, 緯 未 T cells proliferated in umbilical cord blood, but the amplification efficiency of premature infants was significantly lower than that of term infants. Conclusion there are insufficient numbers and functions of 緯 未 T cells in premature infants. This may be an important factor in preterm infants' increased susceptibility to infectious diseases.
