卵清蛋白致敏幼鼠血IL-4、IFN-γ檢測(cè)及意義

【摘要】:目的:通過觀察3周齡SD幼鼠食物過敏(FA)動(dòng)物模型血IL-4、IFN-γ含量及IL-4/IFN-γ比值在致敏前、致敏激發(fā)后變化,探討TH1、TH2型細(xì)胞的活化狀態(tài)在FA發(fā)生中的作用及其意義。 方法:選取16只3周齡清潔級(jí)斷乳雌性SD幼鼠作為實(shí)驗(yàn)對(duì)象,隨機(jī)分為食物過敏組(FA組)和對(duì)照組(NS組),每組8只。通過低劑量卵白蛋白(OVA)腹腔注射基礎(chǔ)致敏及高劑量OVA灌胃激發(fā)建立SD幼鼠OVA食物過敏(FA)。觀察兩組幼鼠致敏前、致敏激發(fā)后體重及活動(dòng)等情況、血OVA-IgE及腸黏膜組織變化進(jìn)行幼鼠FA模型評(píng)價(jià)。采用ELISA法連續(xù)檢測(cè)致敏激發(fā)前后兩組幼鼠血IL-4、IFN-γ含量。 采用SPSS13.0軟件包進(jìn)行統(tǒng)計(jì)分析,α=0.05為顯著性檢驗(yàn)標(biāo)準(zhǔn)。 結(jié)果:FA組幼鼠出現(xiàn)不同程度豎毛,毛發(fā)失去光澤,活動(dòng)減少或俯臥不動(dòng),抓耳撓腮或躁動(dòng)不安及排不成形稀便,與NS組相比,F(xiàn)A組幼鼠末次激發(fā)后體重有減輕趨勢(shì);FA組幼鼠腸道黏膜有較多炎性細(xì)胞和嗜酸性粒細(xì)胞浸潤(rùn);致敏激發(fā)后FA組幼鼠血OVA-IgE含量顯著升高,與NS組比較差別有統(tǒng)計(jì)學(xué)意義(P<0.01)。FA組幼鼠致敏前、致敏激發(fā)后血IL-4含量(ng/L)分別為36.375±4.312、71.075±10.055,NS組分別為35.394±3.635、41.590±6.757,致敏前兩組幼鼠血IL-4含量比較無顯著差別,致敏激發(fā)后FA組IL-4含量明顯升高,與NS組比較差別有統(tǒng)計(jì)學(xué)意義(P<0.01);FA組幼鼠致敏前、致敏激發(fā)后血IFN-γ含量(ng/L)分別為100.849±9.052、259.146±54.008,NS組分別為100.164±18.496、115.654±18.428,,致敏前兩組幼鼠血IFN-γ含量比較無顯著差別,激發(fā)后FA組IFN-γ含量明顯升高,與NS組比較差別有統(tǒng)計(jì)學(xué)意義(P<0.01);隨著致敏后激發(fā)次數(shù)的增多,F(xiàn)A組幼鼠血IL-4、IFN-γ濃度逐漸升高,到末次激發(fā)后達(dá)到高峰,血IL-4、IFN-γ濃度每一次激發(fā)后同一時(shí)間FA組SD幼鼠均較NS組有明顯的升高;兩組幼鼠血清IL-4/IFN-γ比值在致敏前及致敏激發(fā)后比較均無顯著差別(P>0.05)。 結(jié)論:采用OVA腹腔注射基礎(chǔ)致敏,結(jié)合OVA灌胃激發(fā)可建立幼鼠FA動(dòng)物模型;.本課題所建立的FA模型幼鼠TH2及TH1細(xì)胞都處于高功能狀態(tài),可能存在IgE介導(dǎo)的體液免疫反應(yīng)及細(xì)胞介導(dǎo)的遲發(fā)型變態(tài)反應(yīng)。
[Abstract]:Objective: to observe the changes of serum IL-4 / IFN- 緯 content and IL-4 / IFN- 緯 ratio before and after sensitization in a 3-week-old SD rat model of food allergy (FAA), and to explore the role and significance of activation of TH1 / TH2 type cells in the pathogenesis of FA. Methods: sixteen 3-week-old female SD rats with cleanness grade weaning were selected as experimental objects. The rats were randomly divided into two groups: food allergy group (FA group) and control group (NS group, 8 rats in each group). OVA food allergy was induced by intraperitoneal injection of low dose ovalbumin (OVA) and high dose OVA intragastrically. The changes of serum OVA-IgE and intestinal mucosal tissue were evaluated by FA model of young rats. The serum IL-4 and IFN- 緯 levels were continuously detected by ELISA method before and after sensitization. SPSS13.0 software package was used for statistical analysis, 偽 -0. 05 was the significant test standard. Results in the control group, different degrees of hair were observed, the hair lost luster, the activity decreased or the body did not move, the ears scratched the cheek or restless, and the defecation was out of shape. Compared with NS group, the body weight of the young rats in FA group showed a tendency to lose weight after the last stimulation, and there were more inflammatory cells and eosinophil infiltration in intestinal mucosa in FA group, and the serum OVA-IgE content in FA group increased significantly after sensitization and stimulation. There was significant difference between NS group and NS group (P < 0.01). Before sensitization and after sensitization, the serum IL-4 content was 36.375 鹵4.312 ~ 71.075 鹵10.055 鹵10.055N, respectively. There was no significant difference in serum IL-4 content between the two groups before and after sensitization. The IL-4 content of FA group was significantly increased after sensitizing and arousing, and that of NS group was 35.394 鹵3.635U, 41.590 鹵6.757, respectively. There was significant difference between NS group and NS group (P < 0.01). Before and after sensitization, the IFN- 緯 content of serum IFN- 緯 was 100.849 鹵9.052 鹵54.008NS group (100.164 鹵18.496115.654 鹵18.428) respectively. Compared with NS group, the difference was significant (P < 0.01), and the serum IL-4 IFN- 緯 concentration in FA group gradually increased with the increase of sensitizing times, and reached the peak after the last stimulation. The serum IL-4 / IFN- 緯 ratio in FA group was significantly higher than that in NS group at the same time after each stimulation, and there was no significant difference in serum IL-4 / IFN- 緯 ratio between the two groups before and after sensitization (P > 0.05). Conclusion: the FA animal model of young rats can be established by intraperitoneal injection of OVA, combined with the stimulation of OVA. The TH2 and TH1 cells of the model of FA in this study are all in high functional state. There may be humoral immune response mediated by IgE and delayed hypersensitivity mediated by cells.
