FoxO1在新疆維吾爾族先天性馬蹄內(nèi)翻足的表達(dá)及其臨床意義

【摘要】:目的探討FoxO1基因在新疆維吾爾族先天性馬蹄內(nèi)翻足(ICTEV)中的表達(dá)及FoxO1與先天性馬蹄內(nèi)翻足畸形程度的相關(guān)性。方法運(yùn)用Western Blot方法檢測(cè)新疆地區(qū)51例維吾爾族先天性馬蹄內(nèi)翻足患兒和25例維吾爾族健康兒童足部肌肉組織中FoxO1蛋白表達(dá)水平;應(yīng)用實(shí)時(shí)熒光定量PCR測(cè)定不同畸形程度維吾爾族先天性馬蹄內(nèi)翻足患兒FoxO1mRNA相對(duì)表達(dá)量。結(jié)果 Western Blot檢測(cè)結(jié)果顯示ICTEV患兒FoxO1蛋白表達(dá)水平(0.52±0.23)低于正常兒童(1.61±0.15),兩組比較差異有統(tǒng)計(jì)學(xué)意義(P=0.017)。實(shí)時(shí)熒光定量PCR檢測(cè)顯示,DimeglioⅡ、Ⅲ、Ⅳ型ICTEV的FoxO1mRNA相對(duì)表達(dá)量分別為1.02±0.24、0.67±0.15、0.24±0.19;維吾爾族ICTEV畸形程度與FoxO1mRNA相對(duì)表達(dá)量呈負(fù)相關(guān)性(P=0.039)。結(jié)論 FoxO1基因轉(zhuǎn)錄水平隨ICTEV畸形程度的加重而逐漸降低,提示FoxO1基因可能參與了維吾爾族ICTEV的整個(gè)發(fā)病過(guò)程。
[Abstract]:Objective to investigate the expression of FoxO1 gene in congenital equinovarus equinovarus in Xinjiang Uygur and the correlation between FoxO1 and the degree of congenital equinovarus deformity. Methods Western Blot method was used to detect 51 cases of congenital equinovarus deformity of Uygur nationality in Xinjiang.
