新生大鼠缺氧缺血性腦損傷HIF-1α表達(dá)與細(xì)胞凋亡的關(guān)系及干預(yù)研究

【摘要】:目的觀察新生大鼠缺氧缺血性腦損傷(HIBD)后及2-甲氧基雌二醇(2ME2)干預(yù)后對(duì)腦組織缺氧誘導(dǎo)因子-1α(HIF-1α)表達(dá)趨勢(shì)的變化與細(xì)胞凋亡的關(guān)系,探討2ME2對(duì)HIBD的影響及可能的作用。方法將7d齡Wistar大鼠120只,隨機(jī)分為假手術(shù)組、HIBD組及2ME2干預(yù)組。其中后兩組又根據(jù)HIBD后處死動(dòng)物的時(shí)間不同隨機(jī)分為7個(gè)亞組。采用免疫組化的方法對(duì)各組新生大鼠腦組織HIF-1α和Caspase-3的表達(dá)進(jìn)行檢測(cè)及用TUNEL法測(cè)定細(xì)胞凋亡指數(shù)(AI)。結(jié)果 HIF-1α正常情況下低水平表達(dá),HIBD后12h達(dá)高峰,后逐漸降低;Caspase-3表達(dá)于HIBD后6h明顯升高,2d時(shí)達(dá)高峰;細(xì)胞凋亡指數(shù)于HIBD后3h升高,隨著觀察時(shí)間的推移,逐漸增多。2ME2干預(yù)后各時(shí)間點(diǎn)HIF-1α和Caspase-3的表達(dá)水平及細(xì)胞凋亡指數(shù)均較HIBD組明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 HIBD后HIF-1α表達(dá)增高,誘導(dǎo)細(xì)胞凋亡,促進(jìn)了HIBD的進(jìn)展;2ME2干預(yù)后,抑制了HIF-1α的表達(dá),從而發(fā)揮腦保護(hù)作用。
[Abstract]:Objective to observe the relationship between the changes of HIF-1 偽 HIF-1 偽 expression and apoptosis in neonatal rats after hypoxic-ischemic brain injury (HIBD) and the intervention of 2-methoxyestradiol (2-methoxyestradiol) 2ME2). To investigate the effect of 2ME2 on HIBD and its possible effects methods 120 Wistar rats aged 7 days were selected. The latter two groups were randomly divided into 7 subgroups according to the time of death after HIBD. The expression of HIF-1 偽 and Caspase-3 in brain tissue of newborn rats in each group was detected by immunohistochemical method. Apoptosis index was measured by TUNEL method. Results the expression of HIF-1 偽 reached its peak 12 hours after HIBD. The expression of Caspase-3 increased significantly at 6 h after HIBD and reached the peak at 2 d after HIBD, and the apoptotic index increased at 3 h after HIBD, and increased with the time of observation. The expression level of HIF-1 偽 and Caspase-3 and the apoptosis index were significantly lower than those in HIBD group. Conclusion the expression of HIF-1 偽 increased after HIBD and induced apoptosis, which promoted the progression of HIBD. It inhibited the expression of HIF-1 偽, thus exerting the protective effect of brain.
