硫化氫提高腸道微循環(huán)灌注保護新生兒壞死性小腸結(jié)腸炎

【摘要】:目的探討硫化氫(H_2S)對新生SD大鼠壞死性小腸結(jié)腸炎(NEC)的腸道微循環(huán)作用。方法出生12 h內(nèi)的新生SD大鼠10只僅用于檢測H_2S合成酶表達(dá),另80只隨機分為4組:正常對照組、NEC模型組、NEC+GYY4137組、NEC+AOAA組。各組分別予以腹腔注射生理鹽水、LPS+生理鹽水、LPS+GYY4137、LPS+AOAA。通過人工喂養(yǎng)-缺氧-冷刺激-脂多糖(LPS)方法復(fù)制新生鼠NEC模型。HE染色、病理評分以及動物顯微鏡拍實體照片顯示腸組織損傷程度,并統(tǒng)計分析各組新生鼠生存時間,采用FLPI2激光散斑血流實時成像系統(tǒng)檢測各組血流量變化,探討H_2S對新生鼠NEC的腸道微循環(huán)保護作用。結(jié)果與模型組比較,當(dāng)給與NEC處理鼠GYY4137時其腸組織血流量升高(P0.05),病理評分降低(P0.05);而給予AOAA時其病理評分較NEC組增加(P0.05),但腸組織血流量差異并無統(tǒng)計學(xué)意義;NEC組較正常組病理評分增高,血流量降低。末端回腸與全腸組織血流量均值差異無統(tǒng)計學(xué)意義。結(jié)論 H_2S(7.5 mg/kg·d)可通過提高腸道微循環(huán)血流量保護NEC,提高NEC新生鼠生存率。
[Abstract]:Objective to investigate the effect of H2S on intestinal microcirculation in neonatal SD rats with necrotizing enterocolitis. Methods Ten neonatal SD rats within 12 hours were used only to detect the expression of H2S synthase. The other 80 rats were randomly divided into 4 groups: normal control group, GYY4137 group, GYY4137 group, intraperitoneal injection of LPS-LPS-GYY4137LPS-AOAA, which were induced by artificial feeding, hypoxia-cold stimulation and lipopolysaccharide (LPS). Neonatal rat NEC model. Pathological scores and animal microscopic images showed the degree of intestinal tissue injury. The survival time of newborn rats in each group was statistically analyzed. The changes of blood flow in each group were detected by FLPI2 laser speckle flow real time imaging system. To investigate the protective effect of H2s on intestinal microcirculation in neonatal rats with NEC. When NEC was given to GYY4137, the intestinal blood flow increased and the pathological score decreased, but the pathological score of AOAA increased than that of NEC group, but there was no significant difference in intestinal blood flow between NEC group and normal group. There was no significant difference in mean blood flow between terminal ileum and total intestinal tissue. Conclusion H2SN 7.5 mg/kg 路d) can improve survival rate of neonatal NEC rats by increasing intestinal microcirculation blood flow.
