七味白術(shù)散治療小兒輪狀病毒腸炎的腸道宏基因?qū)W相關(guān)研究

【摘要】:目的: 利用宏基因組學(xué)方法,通過(guò)動(dòng)物及臨床實(shí)驗(yàn),研究七味白術(shù)散治療輪狀病毒(RV)腸炎中對(duì)腸道宏基因組的影響,以利于探索腸道微生態(tài)在中藥治療輪狀病毒腸炎中的可能作用。 方法: 本項(xiàng)研究包括動(dòng)物實(shí)驗(yàn)及臨床實(shí)驗(yàn)兩階段。在動(dòng)物實(shí)驗(yàn)階段中,選擇5日齡昆明小鼠為研究對(duì)象,按窩別隨機(jī)分為七味白術(shù)散組、模型對(duì)照組及健康對(duì)照組;七味白術(shù)散組及模型對(duì)照組均以輪狀病毒(RV)懸液經(jīng)口感染,造模成功后七味白術(shù)散組以七味白術(shù)散復(fù)方煎液經(jīng)口灌胃,模型對(duì)照組及健康對(duì)照組則喂以等量的生理鹽水;3日后采集各組乳鼠的結(jié)腸內(nèi)容物標(biāo)本,并應(yīng)用宏基因組學(xué)方法,檢測(cè)及對(duì)比其腸道菌群結(jié)構(gòu)。在臨床實(shí)驗(yàn)階段中,選取1例辨證為脾虛證的12個(gè)月大的輪狀病毒腸炎患兒,應(yīng)用宏基因組學(xué)方法,檢測(cè)及對(duì)比其服用七味白術(shù)散前后腸道菌群的變化。 結(jié)果: 動(dòng)物和臨床實(shí)驗(yàn)結(jié)果顯示,RV感染后的乳鼠和嬰兒之間,其腸道菌群結(jié)構(gòu)差異很大,例如,在門(mén)的水平上,前者以變形菌(Proteobacteria)為主,而后者則以放線(xiàn)菌(Actinobacteria)為主。七味白術(shù)散治療后的RV感染乳鼠與模型對(duì)照組、健康對(duì)照組之間,以及七味白術(shù)散治療后的RV感染嬰兒前后,其腸道菌群結(jié)構(gòu)均存在顯著不同。對(duì)選擇的腸道關(guān)鍵菌群——雙歧桿菌和乳桿菌,在動(dòng)物實(shí)驗(yàn)中,七味白術(shù)散組的腸道雙歧桿菌含量高于模型對(duì)照組,而乳桿菌含量則顯著低于模型對(duì)照組;然而,對(duì)RV感染嬰兒,其腸道雙歧桿菌含量在七味白術(shù)散治療后顯著下降,而乳桿菌含量則在治療后顯著增高。 結(jié)論: 七味白術(shù)散的治療改變了RV感染乳鼠和嬰兒的腸道菌群結(jié)構(gòu),但很可能并不是通過(guò)影響單一特定菌群來(lái)起作用;另一方面,由于乳鼠和嬰兒之間的腸道菌群結(jié)構(gòu)差異很大,功能基因組學(xué)的分析可能更有價(jià)值。
[Abstract]:Objective: The effects of Qiwei Baizhu Powder on intestinal macrogenome in the treatment of rotavirus RV) enteritis were studied by means of macrogenomics and animal and clinical experiments. In order to explore the role of intestinal microecology in the treatment of rotavirus enteritis. Methods: This study includes two stages: animal experiment and clinical experiment. In the animal experiment stage, the 5-day-old Kunming mice were selected as the research objects, and randomly divided into Qiwei Baizhu powder group, model control group and healthy control group according to their nest; Qiwei Baizhu Powder group and model control group were infected by rotavirus RV) suspension. After the successful establishment of the model, Qiwei Baizhu Powder group was orally perfused with Qiwei Baizhu Powder compound decoction. The model control group and the healthy control group were fed with the same amount of normal saline. 3 the colonic contents of each group of neonatal rats were collected, and the intestinal microflora structure was detected and compared by means of macrogenomics. A case of rotavirus enteritis aged 12 months with spleen deficiency syndrome was selected to detect and compare the changes of intestinal flora before and after taking Qiwei Baizhu Powder by macrogenomics. Results: Animal and clinical trials have shown that the intestinal flora structure varies significantly between newborn mice and infants infected with RV, for example, at the portal level. The former is Proteobacteria (Proteobacteria). The latter was mainly actinobacteriae). After treatment with Qiwei Atractylodes powder, there were three groups: model control group, model control group and healthy control group. The intestinal flora structure of infants infected with RV after treatment with Qiwei Baizhu Powder was significantly different. The key intestinal flora bifidobacterium and Lactobacillus were selected in animal experiment. The intestinal bifidobacterium content in Qiwei Baizhu Powder group was higher than that in the model control group, while the Lactobacillus content was significantly lower than that in the model control group. However, for infants infected with RV, the intestinal bifidobacterium content decreased significantly after treatment with Qiwei Baizhu Powder, while the content of Lactobacillus increased significantly after treatment. Conclusion: The treatment of Qiwei Baizhu Powder changed the intestinal microflora structure of newborn mice and infants infected with RV, but probably not by affecting a single specific flora. On the other hand, the analysis of functional genomics may be more valuable because of the large differences in intestinal flora structure between newborn mice and infants.
