捕食性真菌Duddingtonia flagrans基因組DNA的提取及基因組survey分析

【摘要】:采用透析膜培養(yǎng)法結(jié)合DNA柱式提取法進(jìn)行基因組DNA提取,同時(shí)利用第2代高通量測(cè)序技術(shù),分析了捕食性真菌Duddingtonia flagrans基因組概況。結(jié)果顯示:透析膜培養(yǎng)結(jié)合DNA柱式提取法所提取的基因組DNA質(zhì)量及濃度均能達(dá)到測(cè)序標(biāo)準(zhǔn);捕食性真菌Duddingtonia flagrans基因組約為37.6 Mb,GC含量為44.95%,基因組雜合度較低。結(jié)果表明:透析膜培養(yǎng)法結(jié)合柱式法提取其基因組DNA的方法簡(jiǎn)便可行,為絲狀真菌DNA提取,提供了新的思路;基因組survey分析為捕食性真菌Duddingtonia flagrans全基因組精細(xì)圖譜的繪制奠定了基礎(chǔ)。
[Abstract]:The genomic DNA was extracted by dialysis membrane culture combined with DNA column extraction, and the genomic profile of predatory fungi Duddingtonia flagrans was analyzed by the second generation high throughput sequencing technique. The results showed that the quality and concentration of genomic DNA extracted by dialysis membrane culture combined with DNA column extraction could meet the sequencing standard, and the genomic DNA content of predatory fungi Duddingtonia flagrans was about 37.6 Mb,GC and 44.95%, and the heterozygosity of genome was low. The results showed that the method for extracting genomic DNA from filamentous fungi by dialysis membrane culture combined with column method was simple and feasible, which provided a new way to extract DNA from filamentous fungi. Genomic survey analysis laid a foundation for mapping the whole genome fine map of predatory fungi Duddingtonia flagrans.
