PCV2病毒生物礦化條件的優(yōu)化

【摘要】:以礦化效率為評價標(biāo)準(zhǔn),對豬圓環(huán)病毒2型(PCV2)生物礦化的反應(yīng)溫度、反應(yīng)時間及Ca/P比、抗原初始p H、添加劑用量等條件進(jìn)行優(yōu)化,并通過小鼠實驗檢測礦化病毒的免疫原性。結(jié)果顯示,生物礦化最優(yōu)處理條件為初始p H 6.5,Ca/P 1.67,4℃礦化12 h,Mg2+/Ca2+為1.0。在該反應(yīng)條件下,病毒粒子能夠形成球形結(jié)構(gòu)的鈣磷PCV2粒子,粒徑為200~500 nm。不同批次PCV2抗原礦化率皆在90%以上,且該方法可去除大量雜蛋白。小鼠免疫礦化PCV2病毒顆粒后14~28 d,抗體水平與商品苗對照組、未礦化PCV2病毒組基本相同,28 d后對照組抗體水平下降明顯,而PCV2礦化顆粒組的抗體基本維持在原水平,表明PCV2礦化顆粒具有緩釋抗原的作用。
