葡萄籽提取物減緩西雜肉?漳c上皮細胞熱應激反應的功能與機制研究

【摘要】:葡萄籽提取物(Grape seed extract,GSE)是多酚類物質(zhì)的重要來源之一,具有抗氧化、抗炎和免疫調(diào)節(jié)的功能。本試驗旨在研究GSE緩解肉牛空腸上皮細胞(Jejunum epithelial cells,JEC)熱應激反應的作用與機制。在肉牛(西門塔爾♂×秦川牛Q的雜交后代)空腸上皮細胞培養(yǎng)基中分別添加0、10、20、40和80μg/mL的GSE,然后將各濃度梯度下的JEC細胞分別在37℃、40℃、43℃和46℃下進行2h的培養(yǎng)。研究顯示,當細胞培養(yǎng)溫度為40℃時,添加GSE時細胞谷胱甘肽過氧化物酶(GSH-Px)活性(P=0.04)、細胞活力(P=0.004)、細胞表皮生長因子(Epidermal growth factor,EGF;P=0.03)和細胞表皮生長因于受體(Epidermal growth factor receptor,EGFR;P=0.01)mRNA表達均有升高。在同一溫度下,添加GSE降低細胞中自細胞介素(Interleukin 8.IL-8)(P=0.01)、腫瘤壞死因子α(TNF-α)mRNA的表達,同時降低IL-1β(P=0.02)、Toll樣受體4(TLR4;P=0.04)蛋白和熱休克蛋白70(HSP70;P0.001)的濃度。當細胞培養(yǎng)溫度為43℃時,添加GSE組細胞的過氧化氫酶活性(P=0.04)、細胞活力(P0.001).EGF(P0.001)和EGFR(P0.001)mRNA的表達均得到提升,而IL-1β(P0.001).TLR4(P=0.03)和HSP70(P0.001)的濃度以及IL-8(P0.001)、TLR4(P=0.002)和TNF-α(P0.001)的mRNA表達卻顯著降低。T×GSE互作效應在JEC細胞IL-1β(P0.001)、IL-8(P0.001)、TNF-α(P=0.01)、HSP70(P=0.04)的濃度和細胞活力(P0.001)上得到體現(xiàn)。本試驗研究結(jié)果表明,在特定溫度范圍內(nèi),GSE能夠從提高細胞抗氧化能力以及降低炎性反應兩個方面緩解JEC細胞的熱應激反應。
