無(wú)血清條件下UC-MSCs和BMECs共培養(yǎng)對(duì)IGF-Ⅰ表達(dá)的影響

【摘要】:試驗(yàn)旨在探討無(wú)血清條件下臍帶間充質(zhì)干細(xì)胞(UC-MSCs)和奶牛乳腺上皮細(xì)胞(BMECs)共培養(yǎng)對(duì)類(lèi)胰島素樣生長(zhǎng)因子-Ⅰ(IGF-Ⅰ)表達(dá)的影響,將UC-MSCs和BMECs按1∶2直接共培養(yǎng),對(duì)比單純培養(yǎng)的兩種細(xì)胞,48h時(shí)利用ELISA法檢測(cè)各組上清IGF-Ⅰ濃度水平,再利用Transwell小室將兩種細(xì)胞分離培養(yǎng),實(shí)時(shí)熒光定量PCR檢測(cè)各組細(xì)胞IGF-ⅠR、IGF-ⅠmRNA的表達(dá)。結(jié)果顯示,共培養(yǎng)中IGF-Ⅰ濃度顯著高于UC-MSCs組(P0.05),極顯著高于BMECs組(P0.01);UC-MSCs/BMECs、BMECs/UC-MSCs組IGF-ⅠmRNA表達(dá)值均極顯著高于單純培養(yǎng)組(P0.01),BMECs/UC-MSCs組顯著高于UC-MSCs/BMECs組(P0.05);UC-MSCs/BMECs、BMECs/UC-MSCs組IGF-ⅠR mRNA表達(dá)值顯著或極顯著高于單純培養(yǎng)組(P0.05;P0.01),BMECs/UC-MSCs組較UCMSCs/BMECs組差異顯著(P0.05)。綜上,體外無(wú)血清培養(yǎng)條件下,UC-MSCs和BMECs共培養(yǎng)可顯著提高IGF-ⅠR及IGF-ⅠmRNA表達(dá),IGF-Ⅰ濃度水平和IGF-ⅠR mRNA的表達(dá)具有一致性,IGF-Ⅰ主要存在于UCMSCs中。
