小花棘豆中毒對(duì)和田羊母羊繁殖器官的影響

【摘要】:試驗(yàn)旨在探討小花棘豆(Oxytropis glabra DC)中毒對(duì)和田羊母羊繁殖器官指數(shù)、性激素水平和相關(guān)基因mRNA表達(dá)量的影響。以小花棘豆中毒和田羊母羊?yàn)檠芯繉?duì)象,采集血清后屠宰,采集試驗(yàn)羊的丘腦、垂體和卵巢組織,測定其臟器系數(shù),檢測血清中促性腺激素釋放激素(GnRH)、促卵泡素(FSH)、促黃體素(LH)、雌二醇(E2)和孕酮(P4)含量的變化,并檢測各組織中相關(guān)繁殖基因的表達(dá)。結(jié)果顯示,小花棘豆中毒和田羊母羊丘腦、垂體和卵巢指數(shù)均極顯著升高(P0.01),且卵巢表面卵泡數(shù)顯著下降(P0.05),HE染色表明丘腦神經(jīng)元細(xì)胞固縮、濃染;垂體中細(xì)胞核變形、濃染,胞漿減少;卵巢中初級(jí)卵母細(xì)胞溶解、消失,間質(zhì)血管擴(kuò)張。小花棘豆中毒和田羊母羊血清中GnRH、FSH、LH、E2和P4含量均極顯著下降(P0.01),丘腦中Kiss-1、GPR54、ERαmRNA,垂體中GnRHR mRNA和卵巢中FSHR、LHR mRNA表達(dá)量均極顯著下降(P0.01)。結(jié)果表明,小花棘豆毒性成分可通過丘腦—垂體—性腺軸影響和田羊母羊的生殖系統(tǒng)。
[Abstract]:The aim of the experiment was to investigate the effects of (Oxytropis glabra DC) poisoning on reproductive organ index, sex hormone level and mRNA expression of related gene in Hetian sheep. Serum samples were collected and slaughtered, thalamus, pituitary and ovary tissues were collected, organ coefficients were measured, and serum gonadotropin releasing hormone (GnRH),) follicle stimulating hormone (FSH),) was detected. Luteinizing hormone (LH), estradiol (E2) and progesterone (P4) were detected and the expression of related reproductive genes was detected. The results showed that the indexes of thalamus, pituitary and ovary were significantly increased (P0.01), and the number of follicles on the surface of ovary decreased significantly (P0.05), HE staining showed that thalamic neurons were pyknotic and stained intensively). The nuclei of the pituitary gland were deformed, stained and cytoplasm decreased, the primary oocytes in the ovary dissolved and disappeared, and the interstitial blood vessels were dilated. The levels of GnRH,FSH,LH,E2 and P4 in serum decreased significantly (P0.01), and the expression of GnRHR mRNA and FSHR,LHR mRNA in Kiss-1,GPR54,ER 偽 mRNA, pituitary and ovary in thalamus decreased significantly (P0.01). The results showed that the toxic components of Acanthopsis cantonensis could affect the reproductive system of Hetian sheep through the axis of thalamus-pituitary-gonad.
