鴨病毒性肝炎活疫苗的耐熱凍干工藝策略設(shè)計(jì)

【摘要】:【目的】為了提高鴨病毒性肝炎弱毒(DHV)活疫苗的耐熱性能!痉椒ā窟x用蔗糖、普魯蘭糖、明膠、甘油、磷酸鹽等成分設(shè)計(jì)耐熱保護(hù)劑組方,根據(jù)配方的共晶點(diǎn)、塌陷溫度科學(xué)設(shè)計(jì)凍干程序,并比較了不同預(yù)凍方式對(duì)鴨病毒性肝炎活疫苗的效價(jià)影響!窘Y(jié)果】耐熱保護(hù)劑配方A在37℃10 d、15 d病毒損失分別為0.35、0.61 Lg;對(duì)快凍、慢凍以及退火3種不同的凍干方式進(jìn)行比較,發(fā)現(xiàn)對(duì)鴨肝炎病毒耐熱損失由大到小依次為:退火慢凍快凍;疫苗長(zhǎng)期保存期試驗(yàn)結(jié)果表明,可在2~8℃可保存2年;采用10倍劑量免疫2日齡雛鴨均未出現(xiàn)死亡,且精神飲食體重增長(zhǎng)均正常!窘Y(jié)論】采用A配方與快速凍干凍方式的耐熱性能最好,延長(zhǎng)保存期并且提高了耐熱性能,為動(dòng)物疫苗耐熱新劑型的研發(fā)提供技術(shù)支撐。
