復(fù)合益生菌緩解蛋雞熱應(yīng)激效果及機(jī)理研究

【摘要】:近年來,全球動物產(chǎn)業(yè)因熱應(yīng)激已造成了巨大的經(jīng)濟(jì)損失。因此本課題以蛋雞熱應(yīng)激為出發(fā)點,旨在研究屎腸球菌HDRs Ef1和枯草芽胞桿菌HDRa BS1復(fù)合益生菌制劑緩解動物熱應(yīng)激的可行性及其機(jī)制,為新型抗應(yīng)激生物制劑的開發(fā)奠定理論基礎(chǔ)和實踐依據(jù)。本試驗選取856只280日齡產(chǎn)蛋率相近海蘭褐商品蛋雞,隨機(jī)分為3個組:高溫對照組(33℃)、復(fù)合益生菌組[100g/t屎腸球菌(50億CFU/g)+400g/t枯草芽胞桿菌(100億CFU/g)+33℃]、室溫對照組(26℃),每組3個重復(fù),均飼喂玉米-豆粕基礎(chǔ)日糧,試驗期為20 d。試驗結(jié)果及結(jié)論如下:生產(chǎn)性能和雞蛋品質(zhì)方面:與高溫對照組相比,該復(fù)合益生菌組在全期產(chǎn)蛋率、日采食量、平均蛋重方面分別提高了3.99%(P0.01)、6.29%(P0.05)、3.29%(P0.05),但沒有達(dá)到室溫對照組水平,且在料蛋比、腹瀉率、蛋破損率和雞群死亡率等方面均有降低;在蛋殼厚度、強(qiáng)度、蛋白高度、蛋黃顏色和哈氏單位方面分別提高了4.35%(P0.05)、16.72%(P0.05)、15.19%(P0.05)、5.66%(P0.05)和6.56%(P0.05)。血液指標(biāo)方面:與高溫對照組相比,該復(fù)合益生菌組在血清IL-1、內(nèi)毒素、腎上腺素方面分別降低了28.38%(P0.05)、46.07%(P0.05)、31.68%(P0.05);IL-10、孕酮分別提高了137.34%(P0.05)、83.52%(P0.05),而與TNF-α、ND-Ig G相比差異不顯著(P0.05)。腸道屏障功能方面:利用Real-time PCR技術(shù)分析了回、盲腸中特定菌群數(shù)量的變化,通過分析該復(fù)合益生菌顯著降低盲腸大腸桿菌的數(shù)量(P0.05),顯著升高回腸乳酸桿菌的數(shù)量(P0.05),使失調(diào)的腸道菌群組成得到改善;通過Real-time PCR技術(shù)分析該復(fù)合益生菌能極顯著上調(diào)緊密連接蛋白o(hù)ccludin、ZO-1(回、盲腸)和JAM-A(盲腸)基因m RNA的表達(dá)(P0.01),上調(diào)回、盲腸黏膜修復(fù)基因EGF m RNA的表達(dá)(P0.01),上調(diào)回、盲腸黏液蛋白MUC-2基因m RNA的表達(dá)(P0.01;10 d);組織切片結(jié)果分析該復(fù)合益生菌能顯著提高腸絨毛高度(P0.01),而隱窩深度之間無顯著差異(P0.05);Western-blot方法分析該復(fù)合益生菌促進(jìn)免疫蛋白A的表達(dá)和分泌。由此可見,添加該復(fù)合益生菌能改善腸道菌群平衡、修復(fù)腸黏膜、上調(diào)緊密連接蛋白的表達(dá),多層次地增強(qiáng)腸道屏障功能,從而能顯著降低蛋雞血清中內(nèi)毒素和IL-1等核心因子含量,有效緩解蛋雞熱應(yīng)激癥狀,提高其生長性能和雞蛋品質(zhì)。
