高通量測序分析人工養(yǎng)殖成年林麝糞便古菌菌群多樣性

【摘要】:本試驗的目的是采用高通量測序技術(shù)研究人工養(yǎng)殖成年林麝糞便中古菌的結(jié)構(gòu)和組成,并對比不同性別之間的差異。選取12只3歲健康的林麝依據(jù)性別分為雄性組(M組)和雌性組(F組),每組各6只。采集其新鮮糞便,提取DNA,用古菌通用引物PCR擴(kuò)增古菌16S rRNA的V4~V5區(qū),用M i Seq 300 PE測序平臺對擴(kuò)增產(chǎn)物進(jìn)行高通量測序,用QIIM E等軟件對測序序列進(jìn)行分析統(tǒng)計。結(jié)果表明:在從門到屬的各級分類水平上,F組與M組古菌的相對豐度差異均不顯著(P0.05)。M組和F組的組內(nèi)遺傳距離分別為0.16±0.03和0.27±0.06,組間為0.24±0.07,樣品的相似度很高。林麝糞便中的古菌可以分為3個門,優(yōu)勢門為闊古細(xì)菌門(Euryarchaeota);在屬水平上可以分為7個已知屬,優(yōu)勢屬為甲烷短桿菌屬(Methanobrevibacter),其次為熱裸單胞菌屬(Thermogymnomonas)。結(jié)果提示,雄性和雌性林麝糞便中古菌的結(jié)構(gòu)和組成都沒有顯著差異,Methanobrevibacter是林麝糞便中的優(yōu)勢古菌屬
[Abstract]:The purpose of this study was to study the structure and composition of archaic bacteria in the feces of adult musk deer cultured in artificial forest by using high-throughput sequencing technique and to compare the differences between sexes. Twelve healthy forest musk deer aged 3 years were divided into male group (M group) and female group (F group) according to their sex. Fresh feces were collected, DNA, was extracted and the V4~V5 region of 16s rRNA was amplified by PCR, high throughput sequencing was carried out by M i Seq 300 PE sequencing platform, and the sequence was analyzed by QIIM E and other software. The results showed that there was no significant difference in relative abundance between group F and group M at the taxonomic level from phylum to genus (P 0.05). The genetic distances in group). M and group F were 0.16 鹵0.03 and 0.27 鹵0.06, respectively. The similarity between the two groups was 0.24 鹵0.07, and the similarity of the samples was very high. The ancient bacteria in the faeces of musk deer can be divided into 3 phylum, and the dominant phylum is (Euryarchaeota);, which can be divided into 7 known genera at the generic level, and the dominant genus is (Methanobrevibacter), followed by (Thermogymnomonas). The results showed that there was no significant difference in the structure and composition between male and female forest musk deer faeces, and Methanobrevibacter was the dominant ancient fungus in the faeces of forest musk deer.
