牛PPARα、PPARγ基因多態(tài)性及其與生長性狀相關(guān)性分析

【摘要】:肌內(nèi)脂肪含量是影響牛肉品質(zhì)的一個重要因素,它直接影響肉的嫩度和風(fēng)味。過氧化物酶體增殖激活受體PPARα和PPARγ基因是調(diào)控肉牛肌內(nèi)脂肪沉積的相關(guān)基因。牛生長性狀與脂肪沉積之間具有相關(guān)性,本研究以郟縣紅牛、魯西牛、南陽牛、秦川牛、渤海黑牛和青海高原牦牛6個品種共計514頭牛為實(shí)驗(yàn)材料,利用PCR-RFLP結(jié)合DNA池測序等技術(shù),研究了與牛脂肪代謝相關(guān)PPARa、PPARy基因的遺傳多態(tài)性,并對上述多態(tài)位點(diǎn)與郟縣紅牛、魯西牛、南陽牛和秦川牛的多個生長性狀指標(biāo)進(jìn)行了關(guān)聯(lián)分析,為進(jìn)一步闡明相關(guān)基因影響牛經(jīng)濟(jì)性狀的分子機(jī)制提供參考依據(jù)。本研究主要得到以下結(jié)果: 1、牛PPARa基因SNPs檢測及其與肉牛生長性狀相關(guān)性分析 在牛PPARa基因檢測到3個突變位點(diǎn),分別為:第2內(nèi)含子43334bp處A→G突變,第3內(nèi)含子24bp處G→A突變,第8外顯子326bp處T→C突變。在郟縣紅牛、南陽牛、魯西牛和秦川牛四個地方黃牛品種中,PPARα基因43334(A/G)、24(G/A)位點(diǎn)呈現(xiàn)中度多態(tài),326(T/C)位點(diǎn)屬于低度多態(tài)。關(guān)聯(lián)分析顯示:43334(A/G)位點(diǎn)多態(tài)性與魯西牛體斜長、十字部高、腹圍顯著相關(guān)(P0.05);24(G/A)位點(diǎn)多態(tài)性與郟縣紅牛十字部高、坐骨端寬、腰角寬,魯西牛體高、體斜長、十字部高、胸圍、腹圍、體重和重高比,河南地方黃牛坐骨端寬(6月齡)、體重(24月齡)和體斜長(24月齡)指標(biāo)顯著相關(guān)(P0.05);326(T/C)位點(diǎn)多態(tài)性與河南地方黃牛體斜長(6月齡)指標(biāo)顯著相關(guān)(P0.05)。連鎖不平衡分析表明43334(A/G)和24(G/A)位點(diǎn)處于強(qiáng)連鎖狀態(tài),而43334(A/G)與326(T/C)位點(diǎn)之間、24(G/A)與326(T/C)位點(diǎn)之間均為弱連鎖或不連鎖。在不同群體中,單倍型分布存在差異。 2、牛PPARγ基因SNPs檢測及其與肉牛生長性狀相關(guān)性分析 在牛PPARγ基因中檢測到3個突變位點(diǎn),分別為:PPARγ基因第2內(nèi)含子59bp處C→G突變,第7外顯子20bp處C→T突變,第7外顯子50bp處T→C突變。在郟縣紅牛、南陽牛、魯西牛和秦川牛四個地方黃牛品種中,PPARy基因59(C/G)位點(diǎn)呈現(xiàn)中度多態(tài),20(C/T)和50(T/C)位點(diǎn)均屬于低度多態(tài)。關(guān)聯(lián)分析顯示:59(C/G)位點(diǎn)多態(tài)性與郟縣紅牛體重,南陽牛體斜長、腹圍、腰角寬、坐骨端寬和十字部高指標(biāo)顯著相關(guān)(P0.05);20(C/T)位點(diǎn)多態(tài)性與郟縣紅牛體高和十字部高指標(biāo)顯著相關(guān)(P0.05);50(T/C)位點(diǎn)多態(tài)性與南陽牛腰角寬,秦川牛十字部高、重高比指標(biāo)顯著相關(guān)(P0.05)。連鎖不平衡分析顯示,59(C/G)、20(C/T)和50(T/C)三個位點(diǎn)兩兩之間均處于弱連鎖或不連鎖狀態(tài)。單倍型在不同群體中分布存在差異。
[Abstract]:Intramuscular fat content is an important factor affecting beef quality, which directly affects the tenderness and flavor of meat. Peroxisome proliferation-activated receptors (PPAR 偽 and PPAR 緯) are related genes regulating fat deposition in muscle of beef cattle. There was a correlation between growth traits and fat deposition in cattle. In this study, 514 cattle were used as experimental materials, including Jiaxian Red Bull, Luxi cattle, Nanyang cattle, Qinchuan cattle, Bohai Black cattle and Qinghai Plateau Yak. The genetic polymorphisms of PPARa,PPARy gene associated with fat metabolism in cattle were studied by using PCR-RFLP and DNA pool sequencing techniques. The polymorphic loci were compared with Jiaxian red cattle and Luxi cattle. Correlation analysis of several growth traits between Nanyang cattle and Qinchuan cattle was carried out to provide a reference for further elucidating the molecular mechanism of the influence of related genes on cattle economic traits. The main results of this study were as follows: 1. Three mutation sites were detected in bovine PPARa gene by SNPs detection of bovine PPARa gene and its correlation with growth traits of beef cattle. The results were as follows: a / G mutation at intron 2 43334bp; G A mutation in intron 3 24bp and T C mutation in exon 8 326bp. In Jiaxian Red cattle, Nanyang cattle, Luxi cattle and Qinchuan cattle, the PPAR 偽 gene 43334 (A / G), 24 (G / A) loci were moderately polymorphic, and 326 (T / C) loci were low polymorphic. The correlation analysis showed that the polymorphism of 43334 (Ar / G) locus was significantly correlated with the length of the body, the height of the cross and the abdominal circumference of Luxi cattle (P0.05). The polymorphism of 24 (G / A) locus was associated with the height of cross, the width of ischium, the width of waist angle, the height of body, the oblique length of body, the height of cross, chest circumference, abdominal circumference, weight and height of Jiaxian red cattle, and the ischium width of Henan local cattle (6 months old). Body weight (24 months old) and body oblique length (24 months old) were significantly correlated (P0.05); The polymorphism of 326 (T / C) locus was significantly correlated with the body oblique length (6 months old) of Henan local cattle (P0.05). Linkage disequilibrium analysis showed that the 43334 (Ar / G) and 24 (G / A) loci were strongly linked, while 43334 (Ar / G) and 326 (T / C) loci were in a strong linkage state. 24 (G / A) and 326 (T / C) loci were weakly linked or unlinked. The haplotype distribution was different in different populations. 2. SNPs detection of bovine PPAR 緯 gene and its correlation with growth traits of beef cattle revealed three mutation sites in bovine PPAR 緯 gene, namely, C G mutation at 59bp in intron 2 of PPAR 緯 gene and C T mutation at 20bp in exon 7 of PPAR 緯 gene. T-C mutation at exon 7 50bp. In Jiaxian Red cattle, Nanyang cattle, Luxi cattle and Qinchuan cattle, the PPARy gene 59 (C / G) locus was moderately polymorphic, and the 20 (C / T) and 50 (T / C) loci were all low polymorphic. Correlation analysis showed that the polymorphism of 59 (C / G) locus was significantly correlated with Jiaxian red cattle body weight, lean length, abdominal circumference, waist angle width, ischium width and cross height index (P0.05). The 20 (C / T) polymorphism was significantly correlated with the body height and cross height of Jiaxian Red cattle (P0.05), and the 50 (T / C) polymorphism was significantly correlated with the waist angle width of Nanyang cattle, Qinchuan cattle cross height and weight to height ratio (P0.05). Linkage disequilibrium analysis showed that 59 (C / G), 20 (C / T) and 50 (T / C) were weakly linked or unlinked. Haplotypes have different distribution in different populations.
