AMPK在應(yīng)激影響卵母細(xì)胞發(fā)育中的作用

【摘要】:應(yīng)激可以通過兩個方面影響雌性動物的繁殖:(1)干擾排卵;(2)影響植入后的卵母細(xì)胞成熟。這個過程是通過HPA軸和HPG軸的交互作用,影響繁殖的順利進(jìn)行。繁殖受到能量調(diào)控的影響,但調(diào)節(jié)繁殖的多種生理途徑間的關(guān)系尚未完全闡明。促性腺激素釋放激素(GnRH)神經(jīng)元通過整合眾多生理信號從循環(huán)水平調(diào)節(jié)GnRH的釋放,可能會參與調(diào)節(jié)與生殖有關(guān)的能量調(diào)控。GnRH神經(jīng)元可以接收葡萄糖信號,通過AMPK等信號通路進(jìn)行能量調(diào)控。本研究旨在通過證實AMPK參與卵母細(xì)胞成熟,AMPK和糖皮質(zhì)激素有交互影響的作用。最終證明應(yīng)激條件下,AMPK通路會參與母豬卵母細(xì)胞發(fā)育。建立卵母細(xì)胞體外培養(yǎng)體系,配制母豬卵母細(xì)胞成熟培養(yǎng)液。在39℃,CO2濃度5%和飽和濕度的條件下進(jìn)行培養(yǎng)。使用AMPK激活劑(AICAR)和抑制劑(Compound C)分別處理卵母細(xì)胞,統(tǒng)計卵母細(xì)胞成熟率;使用糖皮質(zhì)激素處理卵母細(xì)胞,統(tǒng)計卵母細(xì)胞成熟率;在添加糖皮質(zhì)激素處理細(xì)胞的同時,分別激活和阻斷AMPK信號,檢測卵母細(xì)胞成熟率的變化。建立小鼠熱應(yīng)激模型,對同期發(fā)情并超數(shù)排卵處理的小鼠熱應(yīng)激,檢測熱應(yīng)激對小鼠繁殖系統(tǒng)及下丘腦AMPK通路、血液指標(biāo)的影響,使用藥物(膽汁酸)對其的治療作用。進(jìn)行了兩次關(guān)于母豬繁殖力的省內(nèi)調(diào)研:第一次以調(diào)查問卷形式進(jìn)行,目的是了解當(dāng)前省內(nèi)母豬養(yǎng)殖現(xiàn)狀和母豬繁殖力概況,一次以實地調(diào)研形式對省內(nèi)養(yǎng)殖集團(tuán)下屬養(yǎng)殖場進(jìn)行調(diào)研,目的是通過調(diào)研,搞清環(huán)境中對母豬繁殖力造成影響的應(yīng)激因素并提出解決建議。
[Abstract]:Stress can affect the reproduction of female animals in two ways: (1) interfere with ovulation and (2) affect oocyte maturation after implantation. This process is affected by the interaction between HPA axis and HPG axis. Reproduction is affected by energy regulation, but the relationship between many physiological pathways regulating reproduction has not been fully clarified. Gonadotropin releasing hormone (GnRH) neurons regulate the release of GnRH from the circulatory level by integrating many physiological signals, which may be involved in regulating the energy regulation associated with reproduction. GnRH neurons can receive glucose signals. The energy is regulated by signal pathways such as AMPK. The aim of this study was to demonstrate that AMPK is involved in oocyte maturation and that AMPK and glucocorticoid play an interactive role in oocyte maturation. Finally, it was proved that AMPK pathway would participate in oocyte development in sows under stress.
