絨山羊皮膚毛囊發(fā)育不同時期初級、次級毛囊比較轉錄組學研究
[Abstract]:Inner Mongolia cashmere goats are valuable germplasm genetic resources, so it is very important to study the mechanism of cashmere development and to search for important candidate genes related to the growth of cashmere in Inner Mongolia Cashmere Goat (Inner Mongolia Cashmere Goat). High throughput sequencing technique was used to select 3 RNA samples of primary hair follicles and secondary hair follicles of three Inner Mongolia white cashmere goats at different stages (growth period November, retreat period January, rest month March). The results were analyzed by bioinformatics and qRT-PCR. The results showed that: 1, 9 groups of differential genes were obtained from the primary and secondary hair follicles of three cashmere goats at different growth stages. GO and Pathway analysis of differentially expressed genes were carried out. GO analysis showed that most of the items significantly enriched were associated with extracellular matrix, ribosomal structure, cell translocation and elongation, and cytoplasmic small ribosome. Pathway analysis showed that differential genes mainly affect ribosomal metabolism, amino acid metabolism, vitamin metabolism, sulfur metabolism, neomycin biosynthesis and other functions. Different genes were screened in the primary and secondary hair follicles of cashmere goats at different growth stages. It was found that all of these genes had eight continuous trend changes, respectively,: down down: up down: down up up: up down dowm;. Up down up: up up; At the beginning, the first three trends in the secondary hair follicle group were significant. The analysis of the differentially expressed genes of these eight trends will provide a powerful basis for digging out the mechanism of periodic growth regulation of cashmere goat hair follicles (especially secondary hair follicles). The transcriptome sequencing of LGR5 and SOSTDC1 genes in wool follicles of cashmere goats in November and March were verified by qRT-PCR technique. The results showed that the relative expression of LGR5.SOSTDC1 in wool follicles of cashmere goats in November was significantly higher than that in March. This is basically consistent with the sequencing results. The results show that the sequencing results are true and effective.
