【摘要】：對(duì)綠色產(chǎn)色鏈霉菌生產(chǎn)阿維拉霉素的發(fā)酵條件進(jìn)行優(yōu)化。在單因素試驗(yàn)的基礎(chǔ)上,用Plackett-Burman試驗(yàn)設(shè)計(jì)篩選影響阿維拉霉素發(fā)酵的重要參數(shù)。先采用最陡爬坡試驗(yàn)逼近阿維拉霉素最大生產(chǎn)區(qū)域,再利用Box-Behnken設(shè)計(jì)確定重要參數(shù)的最佳水平。試驗(yàn)結(jié)果表明:油酸添加量、油酸添加時(shí)間、培養(yǎng)基初始p H值對(duì)阿維拉霉素的合成影響顯著。通過(guò)響應(yīng)面分析得到一元二階方程,對(duì)方程求解得到優(yōu)化的參數(shù)值,即油酸添加量0.43%,添加時(shí)間71.28 h,培養(yǎng)基初始p H 6.98。在該參數(shù)值下進(jìn)行阿維拉霉素發(fā)酵,阿維拉霉素產(chǎn)量達(dá)1 207.76μg/m L,較對(duì)照提高了109.97%,與模型預(yù)測(cè)值差異不顯著,說(shuō)明建立的模型是有效的。
[Abstract]:The fermentation conditions for the production of Avila mycin by streptomyces green were optimized. On the basis of single factor experiment, the important parameters affecting the fermentation of Avila mycin were selected by Plackett-Burman test design. The steepest climbing test was used to approximate the maximum production area of Avila mycin, and then the optimal level of important parameters was determined by Box-Behnken design. The results showed that the amount of oleic acid, the time of oleic acid addition and the initial pH value of culture medium had significant effects on the synthesis of Avila mycin. The second order equation was obtained by response surface analysis, and the optimized parameter value was obtained by solving the equation, that is, the amount of oleic acid was 0.43, the adding time was 71.28 h, and the initial medium pH 6.98. The yield of Avila mycin reached 1 207.76 渭 g / mL, which was 109.97 渭 g / mL higher than that of the control, and there was no significant difference from the predicted value of the model, which indicated that the established model was effective.
