【摘要】：為探討牛種布魯氏菌sigma因子RpoE2的功能,采用酵母雙雜交技術(shù)對RpoE2蛋白與anti-sigma因子之間的互作進行研究;通過環(huán)境應(yīng)激試驗評價了rpoE2缺失株對強氧化環(huán)境、酸性pH、殺菌性陽離子多肽、鐵缺乏環(huán)境的耐受能力;通過比較親本菌株和缺失株在巨噬細胞感染模型和小鼠感染模型中的生存能力,研究了rpoE2基因是否與牛種布魯氏菌毒力相關(guān)。結(jié)果顯示:牛種布魯氏菌的RpoE2蛋白可以與anti-sigma因子相互作用,牛種布魯氏菌rpoE2基因缺失后,對各種應(yīng)激環(huán)境的耐受能力均無影響,并且巨噬細胞和小鼠感染模型中毒力均不降低。牛種布魯氏菌的RpoE2蛋白是一個ECF16家族的sigma因子,并且其與布魯氏菌的毒力無關(guān)。
[Abstract]:In order to study the function of sigma factor RpoE2 in bovine brucella, the interaction between RpoE2 protein and anti-sigma factor was studied by yeast two-hybrid technique. Environmental stress test was used to evaluate the tolerance of rpoE2 deficient strain to strong oxidative environment, acidic pH, bactericidal cationic polypeptide, and iron deficiency. By comparing the viability of parent strain and deletion strain in macrophage infection model and mouse infection model, the relationship between rpoE2 gene and brucella virulence was studied. The results showed that the RpoE2 protein of Brucella bovis could interact with the anti-sigma factor, and the rpoE2 gene deletion of Brucella spp had no effect on the tolerance of various stress environments. The toxicity of macrophages and mice was not decreased. The RpoE2 protein of bovine brucella is a sigma factor of ECF16 family, and it is not related to the virulence of brucella.
