【摘要】：為分析犬p27蛋白在犬乳腺腫瘤組織中的表達情況,本研究構建了原核表達重組質粒pET-p27,進行重組蛋白的表達,并將純化的重組蛋白免疫新西蘭白兔,制備了抗犬p27蛋白多克隆抗體。將制備的犬p27抗體對臨床84例犬乳腺腫瘤組織中的犬p27蛋白表達情況進行了檢測。結果顯示,制備的抗體效價達1∶12 800以上;western blot檢測顯示,該多克隆抗體能夠與重組蛋白發(fā)生特異性反應;免疫組織化學試驗表明,犬p27蛋白在犬惡性乳腺腫瘤組織中表達率低于犬良性乳腺腫瘤組織,提示犬p27的表達水平可能與犬乳腺腫瘤的惡性程度相關,本實驗為進一步研究犬p27蛋白的生物學功能奠定了基礎。
[Abstract]:In order to analyze the expression of canine p27 protein in canine breast tumor tissues, a prokaryotic expression plasmid pET-p27, was constructed to express the recombinant protein, and the purified recombinant protein was immunized with New Zealand white rabbit. A polyclonal antibody against canine p27 protein was prepared. The expression of p27 protein in 84 cases of canine breast neoplasms was detected by using the prepared canine p27 antibody. The results showed that the antibody titer was over 1:12 by; western blot, and the polyclonal antibody could react specifically with the recombinant protein. The expression rate of p27 protein in canine malignant breast tumor is lower than that in benign mammary gland tumor, suggesting that the expression level of p27 may be related to the malignant degree of canine breast tumor. This study laid a foundation for further study on the biological function of canine p27 protein.
