【摘要】：為探討膜性雌激素受體GPER(G protein-coupled estrogen receptor 1)及雌激素核受體ERα、ERβ在綿羊輸卵管上皮細(xì)胞中的表達(dá)和定位,探討三種受體在間情期綿羊輸卵管中的表達(dá)規(guī)律,通過RT-qPCR方法和免疫印跡方法分別檢測了三種受體基因mRNA水平和蛋白水平的表達(dá)差異;運用免疫組化法對輸卵管組織中三種受體基因進行定位并對結(jié)果進行光密度值分析。結(jié)果,輸卵管上皮細(xì)胞中GPER相對表達(dá)量高于ERα、ERβ(P0.01),ERα的相對表達(dá)量高于ERβ(P0.05)。免疫組化結(jié)果顯示,GPER廣泛分布于綿羊輸卵管上皮細(xì)胞中,主要定位于細(xì)胞膜及細(xì)胞質(zhì)。而ERα及ERβ主要分布于綿羊輸卵管上皮細(xì)胞的細(xì)胞核。陽性細(xì)胞光密度分析結(jié)果與熒光定量RT-qPCR的分析結(jié)果相一致。上述結(jié)果表明,間情期綿羊輸卵管上皮細(xì)胞中同時存在GPER、ERα、ERβ三種雌激素受體表達(dá),且GPER的平均光密度、mRNA相對表達(dá)量最高,ERβ的平均光密度、mRNA相對表達(dá)量最低。
[Abstract]:To investigate the expression and localization of membrane estrogen receptor (GPER (G protein-coupled estrogen receptor 1) and estrogen nuclear receptor (ER 偽 er 尾) in ovine oviductal epithelial cells, and to investigate the expression of these receptors in ovine oviduct during interoestrous period. The expression differences of mRNA and protein levels of three receptor genes were detected by RT-qPCR and Western blotting, and the three receptor genes in oviducts were located by immunohistochemical method and the results were analyzed by optical density. The results showed that the relative expression of GPER in oviduct epithelial cells was higher than that in ER 偽 -ER 尾 (P0.01) and ER 尾 (P0.05). Immunohistochemical results showed that GPER was widely distributed in ovine oviduct epithelial cells, mainly located in cell membrane and cytoplasm. However, ER 偽 and ER 尾 mainly distributed in the nuclei of ovine oviduct epithelial cells. The results of optical density analysis of positive cells were consistent with those of fluorescent quantitative RT-qPCR. The results showed that there were three estrogen receptors of GPER,ER 偽 -ER 尾 in oviductal epithelial cells of sheep during interoestrous period, and the relative expression of GPER was the highest and the lowest was the average optical density of ER 尾.
