乳酸菌富硒條件篩選和毒理試驗及在斷奶仔豬生產(chǎn)上的應(yīng)用

【摘要】:在我國目前及未來畜牧業(yè)發(fā)展中占有重要地位的新型飼料添加劑--安全、高效、無污染的綠色添加劑,已逐漸受到海內(nèi)外很多研究者的關(guān)注,并以此為研發(fā)課題進(jìn)行了大量的研究,開發(fā)研究了很多類型的新型飼料添加劑。本試驗以乳酸菌發(fā)酵米糠富硒生產(chǎn)富硒乳酸菌添加劑為基礎(chǔ),對成品添加劑進(jìn)行有關(guān)毒理試驗為前提,開展斷奶仔豬飼養(yǎng)試驗,探究富硒乳酸菌添加劑在斷奶仔豬飼養(yǎng)過程中的飼喂效果,并為其日后在畜禽養(yǎng)殖過程中的實際應(yīng)用效果提供相關(guān)的實踐依據(jù)。本試驗在研究富硒乳酸菌添加劑的基礎(chǔ)上進(jìn)行了三個試驗:1.乳酸菌發(fā)酵米糠富硒條件的篩選試驗用乳酸菌先進(jìn)行純化、擴(kuò)繁培養(yǎng),之后利用培養(yǎng)好的乳酸菌開展發(fā)酵試驗。經(jīng)過一系列的發(fā)酵篩選試驗后,確定了乳酸菌發(fā)酵米糠富硒的最佳條件:米糠添加量34g,培養(yǎng)基裝液量55mL/250mL,接種量9%,種齡8-16h,發(fā)酵時間48h,發(fā)酵溫度37℃,亞硒酸鈉添加量100μg/mL,在此條件下發(fā)酵米糠,乳酸菌的生物量可達(dá)9.94g/L,有機(jī)硒的生產(chǎn)水平為7.46mg/L。2.富硒乳酸菌對小鼠的蓄積性毒性試驗本試驗在急性毒性試驗的基礎(chǔ)上,將40只昆明(KM)小鼠采用完全隨機(jī)的形式進(jìn)行分組,并根據(jù)KM小鼠每天攝入有效硒的量不同確定為4個組,即40mg/kg組、60mg/kg組和90mg/kg組,每組10只KM小鼠,雄鼠、雌鼠各5只,連續(xù)飼養(yǎng)40d,試驗期間觀察KM小鼠每天的健康狀況,同時進(jìn)行樣品的采集工作及后期生長性能、血清抗氧化指標(biāo)、臟器指數(shù)的測定和病理切片的觀察,試驗結(jié)果表明:飼喂不同劑量的富硒乳酸菌制劑能夠增加公鼠和母鼠的體重、采食量,并顯著降低料重比;對于抗氧化方面發(fā)現(xiàn),飼喂適量的富硒乳酸菌制劑可使公鼠和母鼠的血清抗氧化能力增強(qiáng),過量反而會降低。結(jié)合臟器指數(shù)和病理切片來看,結(jié)果發(fā)現(xiàn),40mg/kg組母鼠的臟器發(fā)育情況和病理切片結(jié)果變化最小,60mg/kg和90mg/kg組母鼠肝臟有輕微腫大及顆粒變性現(xiàn)象發(fā)生;對于公鼠而言,60mg/kg組公鼠的臟器發(fā)育和病理切片結(jié)果變化最小。綜合公鼠和母鼠的各項指標(biāo)可以得出,低量的硒制劑對小鼠的生長有一定的促進(jìn)作用;隨著硒制劑劑量的逐漸增加,小鼠的生長發(fā)育開始出現(xiàn)受阻現(xiàn)象,同時對機(jī)體肝臟也造成了明顯的傷害。3.富硒乳酸菌制劑對仔豬生長性能、免疫及抗氧化能力的影響本試驗選用斷奶天數(shù)相同,體況相近且健康的100頭仔豬,隨機(jī)分為5組,分別為對照組、0.3mg/kg 組、0.6mg/kg 組、0.9mg/kg 組、1.2mg/kg 組,每組 4個重復(fù),一個重復(fù)5頭。正式試驗共42天,在正式試驗開始前先進(jìn)行一周左右的預(yù)飼期,試驗期間對仔豬待測的各項指標(biāo)樣品及時進(jìn)行采集,飼養(yǎng)試驗結(jié)束后,對各項指標(biāo)進(jìn)行測定,試驗結(jié)果表明:富硒乳酸菌制劑在仔豬日糧中的加量為0.9mg/kg時,可顯著提高仔豬的生長性能,增強(qiáng)日糧中某些營養(yǎng)成分的表觀消化率,并能夠明顯改善仔豬的腹瀉情況,同時可增強(qiáng)斷奶仔豬的免疫及抗氧化能力。
[Abstract]:Nowadays and in the future, many researchers at home and abroad have paid more and more attention to the new feed additives which play an important role in the development of animal husbandry in our country, such as safe, efficient and non-polluting green additives. A lot of research has been done on this topic, and many kinds of new feed additives have been developed and studied. Based on the production of selenium-enriched Lactobacillus additives from fermented rice bran with selenium-enriched, and on the premise of toxicological test of product additives, the feeding test of weaned piglets was carried out to explore the feeding effect of selenium-enriched Lactobacillus additives in the feeding process of weaned piglets, and to provide relevant practice for its practical application in livestock and poultry breeding in the Based on the study of selenium-enriched lactic acid bacteria additives, three experiments were carried out: 1. Lactobacillus was used to select selenium-enriched fermentation conditions of rice bran. Lactobacillus was purified and cultured first, then fermented with cultured lactic acid bacteria. The optimum conditions of selenium were as follows: 34 g rice bran, 55 mL/250 mL culture medium, 9% inoculum, 8-16 h seed age, 48 h fermentation time, 37 C fermentation temperature, 100 ug/mL sodium selenite. Under these conditions, the biomass of lactic acid bacteria could reach 9.94 g/L, and the production level of organic selenium was 7.46 mg/L.2. On the basis of acute toxicity test, 40 Kunming (KM) mice were divided into four groups randomly. According to the daily intake of Available Selenium in KM mice, there were 40 mg/kg group, 60 mg/kg group and 90 mg/kg group. Each group had 10 KM mice, 5 male mice and 5 female mice. The KM mice were fed continuously for 40 days. The results showed that feeding different doses of selenium-enriched lactobacillus preparation could increase the body weight of male and female rats, feed intake and significantly reduce the feed-to-weight ratio for antioxidant prescription. It was found that the serum antioxidant capacity of male and female rats was enhanced and the excess was decreased by feeding selenium-enriched lactobacillus. According to organ index and pathological section, the changes of organ development and pathological section were the smallest in the 40 mg/kg group, and slight hepatomegaly and granules were found in the 60 mg/kg and 90 mg/kg groups. The results of organ development and pathological section of male mice in the 60mg/kg group showed that the growth of mice was promoted by low dose of selenium, and the growth and development of mice began to be blocked with the increasing dose of selenium. Selenium-enriched lactobacillus preparation on growth performance, immunity and antioxidant capacity of piglets, 100 piglets with the same weaning days, similar and healthy were randomly divided into five groups: control group, 0.3 mg/kg group, 0.6 mg/kg group, 0.9 mg/kg group, 1.2 mg/kg group, each group of four. A total of 5 piglets were duplicated. The formal experiment lasted for 42 days. The pre-feeding period was about a week before the formal experiment. During the experiment, the piglets were collected in time. After the feeding experiment, the indexes were determined. The results showed that the addition of selenium-enriched lactobacillus preparation in the diet of piglets was 0.9mg/kg. It can significantly improve the growth performance of piglets, enhance the apparent digestibility of some nutrients in the diet, and significantly improve the diarrhea of piglets. It can also enhance the immunity and antioxidant capacity of weaned piglets.
