梅花豬胃腸道5-羥色胺、胃饑餓素和胰高血糖素樣肽-1免疫陽性細(xì)胞分布特征研究

【摘要】:為了解中國優(yōu)良地方豬種梅花豬胃腸道內(nèi)分泌細(xì)胞的分布與形態(tài)特征,本試驗選取6頭30日齡斷奶健康梅花仔豬,在相同條件下飼養(yǎng)至65日齡,所有仔豬飼喂相同日糧,自由采食飲水。試驗結(jié)束時,收集仔豬胃腸道各段組織樣品,應(yīng)用免疫組織化學(xué)方法觀察3種胃腸道內(nèi)分泌激素5-羥色胺(5-HT)、胃饑餓素(ghrelin)和胰高血糖素樣肽-1(GLP-1)免疫陽性細(xì)胞的分布及形態(tài)特征,并對陽性細(xì)胞密度進行分析。結(jié)果顯示,梅花豬的腸道不同部位(十二指腸、空腸、回腸和結(jié)腸)均存在5-HT陽性細(xì)胞,主要位于腸上皮組織中,呈錐形、圓形、梭形等形態(tài),可分為開放型和閉合型,其中十二指陽性細(xì)胞腸密度顯著高于其他腸段(P0.05);梅花豬的胃腸道不同部位均有Ghrelin陽性細(xì)胞,陽性細(xì)胞密度也是隨胃腸道走向遞減,胃腺體部密度最高;梅花豬的腸道不同部位均有GLP-1陽性細(xì)胞,陽性細(xì)胞密度沿腸道走向呈拋物線分布,其中回腸數(shù)量最多。本試驗結(jié)果為進一步探究梅花豬胃腸道生理奠定了基礎(chǔ)。
[Abstract]:In order to understand the distribution and morphological characteristics of endocrine cells in gastrointestinal tract of China's fine local pig breeds, six 30 day weaned healthy Plum Piglet piglets were selected and fed to 65 days of age under the same conditions. All piglets were fed the same diet. Free drinking and drinking water. At the end of the experiment, the distribution and morphological characteristics of immunoreactive cells of three gastrointestinal endocrine hormones (5-HT, (ghrelin) and GLP-1) in piglets were observed by immunohistochemical method. The density of positive cells was analyzed. The results showed that there were 5-HT positive cells in different parts of the intestine (duodenum, jejunum, ileum and colon) of the pig, mainly located in the intestinal epithelium, which were conical, round and fusiform, and could be divided into open type and closed type. The intestinal density of positive cells in 12 fingers was significantly higher than that in other intestinal segments (P0.05), and Ghrelin positive cells were found in different parts of the gastrointestinal tract, and the density of positive cells decreased with the trend of gastrointestinal tract, and the density of gastric glands was the highest. GLP-1 positive cells were found in different parts of the intestinal tract of the pig, and the density of the positive cells was parabola along the direction of the intestine, and the number of the ileum was the most. The results of this study laid a foundation for further exploring the gastrointestinal physiology of Plum Plum Pigs.
