飼糧陰陽(yáng)離子差水平對(duì)小鼠血鈣濃度及胃腸道組織鈣結(jié)合蛋白-D9k mRNA相對(duì)表達(dá)水平的影響

【摘要】:本試驗(yàn)在前期建立了動(dòng)物胃腸道鈣代謝相關(guān)基因表達(dá)水平檢測(cè)體系與表達(dá)譜分析的基礎(chǔ)上,旨在進(jìn)一步確定飼糧陰陽(yáng)離子差(DCAD)水平對(duì)動(dòng)物血鈣濃度和胃腸道組織鈣結(jié)合蛋白-D9k(Ca BP-D9k)mRNA相對(duì)表達(dá)水平的影響,為揭示低DCAD水平防治動(dòng)物低血鈣癥的作用機(jī)制提供依據(jù)。將120只昆明小鼠隨機(jī)分為3組,每組40只,自配種前3 d起分別飼喂DCAD水平為+300(高DCAD水平組,HD組)、+150(對(duì)照組,CON組)、-50(低DCAD水平組,LD組)的飼糧。檢測(cè)母鼠產(chǎn)前20 d(-20 d)、產(chǎn)前5 d(-5 d)、產(chǎn)后當(dāng)天(0 d)、產(chǎn)后3 d(+3 d)血鈣濃度和胃腸道組織Ca BP-D9k mRNA相對(duì)表達(dá)水平。結(jié)果表明,與HD組相比,LD組顯著提高了圍產(chǎn)期內(nèi)0 d、+3 d小鼠血鈣濃度(P0.05),顯著上調(diào)了小腸腸段(十二指腸、空腸、回腸)與結(jié)腸Ca BP-D9k mRNA相對(duì)表達(dá)水平(P0.05),這一效應(yīng)在-5 d、0 d表現(xiàn)最為顯著(P0.05),并在+3 d提高了小鼠空腸與結(jié)腸的Ca BP-D9k mRNA相對(duì)表達(dá)水平(P0.05)。統(tǒng)計(jì)結(jié)果顯示,十二指腸、空腸與結(jié)腸3個(gè)位點(diǎn)的Ca BP-D9k mRNA相對(duì)表達(dá)水平與DCAD水平、血鈣濃度具有顯著的關(guān)聯(lián)性(P0.05)。由此可見(jiàn),降低DCAD水平可上調(diào)動(dòng)物小腸及結(jié)腸腸段Ca BP-D9k mRNA相對(duì)表達(dá)水平,同時(shí)伴隨更高的血鈣濃度。這可能是低DCAD水平有效維持動(dòng)物圍產(chǎn)期血鈣穩(wěn)恒,降低低血鈣發(fā)生率的重要途徑。
[Abstract]:Based on the establishment of an animal gastrointestinal calcium metabolism-related gene expression system and analysis of expression profiles, this study aimed to further determine the effects of dietary anion-cation difference (DCAD) levels on serum calcium concentration and the relative expression of calcium-binding protein-D9k (Ca BP-D9k) mRNA in gastrointestinal tissues, and to reveal the prevention of low DCAD levels. 120 Kunming mice were randomly divided into 3 groups, 40 mice in each group. The levels of DCAD were + 300 (high DCAD level group, HD group), + 150 (control group, CON group), - 50 (low DCAD level group, LD group). The maternal rats were detected 20 days before delivery (- 20 days), 5 days before delivery (- 5 days), on the same day after delivery (0 days). The results showed that compared with HD group, LD group significantly increased the serum calcium concentration (P 0.05) during perinatal period, and significantly increased the relative expression level of Ca BP-D9k mRNA in small intestine (duodenum, jejunum, ileum) and colon (P 0.05), the effect was in - 5 d, 0 d table. The relative expression level of CaBP-D9k mRNA in the jejunum and colon of mice was significantly increased at +3 D (P 0.05). Statistical results showed that the relative expression level of CaBP-D9k mRNA in the duodenum, jejunum and colon was significantly correlated with the level of DCAD and the level of serum calcium (P 0.05). The relative expression of Ca BP-D9k mRNA in the small intestine and colon was accompanied by higher serum calcium concentration, which may be an important way to maintain the perinatal calcium homeostasis and reduce the incidence of hypocalcemia.
