【摘要】：【目的】建立一種能夠區(qū)分豬繁殖與呼吸綜合征病毒(PRRSV)經典株和變異株的RT-nPCR診斷方法,能夠從病料組織、血清或精液中直接進行目的基因檢測,達到快速診斷的目的�！痉椒ā扛鶕�(jù)GenBank上發(fā)表的豬繁殖與呼吸綜合征病毒基因序列,設計并合成了2對引物,通過改變不同的反應體系和反應條件,建立PRRSV經典株和變異株的RT-nPCR診斷方法,并通過靈敏性試驗、特異性試驗、病料組織和血清或精液樣品檢測,驗證建立RT-nPCR方法的適用性�！窘Y果】建立了PRRSV經典株和變異株的RT-nPCR診斷方法,該方法檢測的cDNA含量極限為1.3×10-7 pg/μL;且僅能從PRRSV毒株中擴增到目的條帶,從CH-1R經典株擴增出649bp條帶,從SXXY/2013變異株擴增出562bp條帶,電泳后能清晰區(qū)別,而從其他參考毒株中均不能擴增出目的條帶。從48份疑似病料組織和血清中能直接檢測出31份陽性結果。【結論】建立了一種靈敏度高、特異性好、可直接從病料組織和血清中快速鑒別PRRSV經典株和變異株的RT-nPCR方法。
[Abstract]:[objective] to establish a method for the diagnosis of porcine reproductive and respiratory syndrome virus (PRRSV) (PRRSV) classic strain and variant strain by RT-nPCR, which can detect the target gene directly from the diseased tissue, serum or semen. [methods] according to the gene sequence of porcine reproductive and respiratory syndrome virus published on GenBank, two pairs of primers were designed and synthesized. To establish a RT-nPCR diagnostic method for classical and variant strains of PRRSV, and to detect the tissues and serum or semen samples of PRRSV by sensitivity test, specificity test. [results] A RT-nPCR diagnostic method for classical and variant strains of PRRSV was established. The limit of cDNA content detected by this method was 1.3 脳 10 ~ (-7) pg/ 渭 L, and only the target bands could be amplified from PRRSV strains and 649bp bands could be amplified from CH-1R strains. The 562bp bands were amplified from the SXXY/2013 mutant strains, but the target bands could not be amplified from other reference strains. 31 positive results could be directly detected from 48 samples of suspected samples and sera. [conclusion] A RT-nPCR method with high sensitivity and good specificity was established for the rapid identification of classical and variant strains of PRRSV from diseased tissue and serum.
