【摘要】：為了研究肉雞腹水綜合征(ascites syndrome,AS)肉雞的肺組織、肺動(dòng)脈血管壁結(jié)構(gòu)重塑、肺主動(dòng)脈張力的變化與規(guī)范瞬時(shí)受體電位離子通道(canonical transient receptor potential channel,TRPC)的mRNA表達(dá)情況之間的關(guān)系,采用靜脈注射纖維素顆粒的方法誘發(fā)肉雞腹水征。然后,采用HE染色和Weiger套Van-Giesen彈力纖維染色法觀察肺組織結(jié)構(gòu)變化,Wire-myograph系統(tǒng)測(cè)量KCl刺激后血管張力的變化,RT-PCR方法檢測(cè)肺組織中TRPC mRNA的表達(dá)。結(jié)果顯示:靜脈注射纖維素顆粒誘導(dǎo)的腹水肉雞肺組織間隙增寬,肺實(shí)質(zhì)炎性細(xì)胞浸潤(rùn)及肺動(dòng)脈血管壁結(jié)構(gòu)重塑,且血管張力下降,但是TRPC mRNA在肺組織中的表達(dá)沒有發(fā)生明顯變化。研究結(jié)果表明,肉雞腹水綜合征所引起肺動(dòng)脈血管壁結(jié)構(gòu)重塑以及肺動(dòng)脈張力的變化與TRPC mRNA的表達(dá)無關(guān)。
[Abstract]:In order to study the relationship between lung tissue, pulmonary artery wall remodeling, changes of pulmonary aortic tension and mRNA expression of normal transient receptor potential ion channel (canonical transient receptor potential channel,TRPC) in broilers with ascites syndrome (ascites syndrome,AS). Ascites were induced by intravenous injection of cellulose granules in broilers. Then, the changes of lung tissue structure were observed by HE staining and Van-Giesen elastic fiber staining with Weiger. The changes of vascular tension after KCl stimulation were measured by Wire-myograph system. RT-PCR was used to detect the expression of TRPC mRNA in lung tissue. The results showed that the interstitial space was widened, inflammatory cell infiltration of pulmonary parenchyma and remodeling of pulmonary artery wall were induced by intravenously injected cellulose granules in broilers with ascites, and the vascular tension was decreased. However, there was no significant change in the expression of TRPC mRNA in lung tissue. The results showed that the remodeling of pulmonary artery wall and the change of pulmonary artery tension were not related to the expression of TRPC mRNA in broiler ascites syndrome.
