【摘要】：目的研究吡喹酮注射液在水牛體內(nèi)的藥物動(dòng)力學(xué)特征及其與吡喹酮片之間的相對(duì)生物利用度。方法選用6頭健康成年水牛,采用兩周期隨機(jī)交叉實(shí)驗(yàn)設(shè)計(jì),單劑量?jī)?nèi)服吡喹酮片(20 mg/kg)或肌注吡喹酮注射液(10 mg/kg),高效液相色譜法(HPLC)測(cè)定水牛血漿中吡喹酮濃度,進(jìn)行非房室模型分析,計(jì)算藥動(dòng)學(xué)參數(shù)。結(jié)果水牛內(nèi)服吡喹酮片的峰時(shí)(T_(max))為(0.60±0.29)h,峰濃度(C_(max))為(0.57±0.37)μg/ml,消除半衰期(T_(1/2β))為(0.70±0.42)h,藥-時(shí)曲線下面積(AUC)為(0.80±0.70)(μg/ml)·h。水牛肌注吡喹酮注射液的T_(max)為(0.65±0.49)h,C_(max)為(3.82±1.17)μg/ml,T_(1/2β)為(1.00±0.73)h,AUC為(1.61±0.89)(μg/ml)·h。吡喹酮注射液相對(duì)于吡喹酮片的生物利用度為402.5%。結(jié)論吡喹酮注射液肌注給藥后在水牛體內(nèi)具有吸收迅速,生物利用度高,體內(nèi)分布廣的藥物動(dòng)力學(xué)特征,臨床推薦給藥劑量為10 mg/kg。
[Abstract]:Objective to study the pharmacokinetic characteristics of praziquantel injection in buffalo and its relative bioavailability with praziquantel tablets. Methods six healthy adult buffaloes were used in a two-cycle randomized crossover design. Praziquantel tablets (20 mg/kg) or praziquantel injection (10 mg/kg) were administered intramuscularly. The concentration of praziquantel in buffalo plasma was determined by high performance liquid chromatography (HPLC). The non-atrioventricular model was analyzed and pharmacokinetic parameters were calculated. Results the peak time (T _ (max) of praziquantel tablet was (0.60 鹵0.29) h, the peak concentration (C _ (max) was (0.57 鹵0.37) 渭 g / ml, the elimination half life (T _ (1 / 2 尾) was (0.70 鹵0.42) h, and the area under the drug-time curve was (0.80 鹵0.70) (渭 g/ml) h. The T _ (max) of praziquantel injection was (0.65 鹵0.49) (max) (3.82 鹵1.17) 渭 g 路ml ~ (-1) T _ (1 / 2 尾) was (1.00 鹵0.73) HPA = (1.61 鹵0.89) (渭 g/ml) h. The bioavailability of praziquantel injection was 402.5. Conclusion praziquantel injection has the characteristics of rapid absorption, high bioavailability and wide distribution in the body of buffalo after intramuscular injection. The recommended dosage of praziquantel injection is 10 mg / kg.
