bta-miR-205和bta-miR-497調(diào)控牛病毒性腹瀉病毒復(fù)制的研究

【摘要】:為探究bta-miR-205和bta-miR-497對(duì)牛病毒性腹瀉病毒(BVDV)復(fù)制的調(diào)控作用,在前期建立BVDV感染MDBK細(xì)胞miRNAs差異表達(dá)譜的基礎(chǔ)上,篩選出表達(dá)量顯著差異的bta-miR-205和btamiR-497,通過生物信息學(xué)軟件預(yù)測(cè)、熒光素酶報(bào)告基因檢測(cè)等方法鑒定bta-miR-205和bta-miR-497的靶基因。應(yīng)用RT-qPCR和病毒滴度測(cè)定等方法驗(yàn)證bta-miR-205和bta-miR-497對(duì)BVDV復(fù)制的影響。結(jié)果發(fā)現(xiàn),bta-miR-205能顯著抑制BVDV的復(fù)制,而bta-miR-497顯著促進(jìn)BVDV的復(fù)制。研究結(jié)果為BVD防控提供了新的思路和依據(jù)。
[Abstract]:In order to investigate the regulatory effect of bta-miR-205 and bta-miR-497 on (BVDV) replication of bovine viral diarrhea virus (BVDV), bta-miR-205 and btamiR-497were screened out on the basis of miRNAs differential expression profiles of MDBK cells infected with BVDV in the early stage, and predicted by bioinformatics software. Luciferase reporter gene detection was used to identify the target genes of bta-miR-205 and bta-miR-497. The effects of bta-miR-205 and bta-miR-497 on BVDV replication were tested by RT-qPCR and virus titer assay. The results showed that bta-miR-205 could significantly inhibit the replication of BVDV, while bta-miR-497 significantly promoted the replication of BVDV. The results provide a new idea and basis for the prevention and control of BVD.
