五加芪水煎液對(duì)小鼠和雛雞免疫功能的影響

【摘要】:本試驗(yàn)以研究五加芪水煎液分別對(duì)小鼠和雛雞免疫功能的作用為目的。試驗(yàn)由體外試驗(yàn)和體內(nèi)試驗(yàn)兩部分組成。體外試驗(yàn):小鼠脾臟淋巴細(xì)胞與不同濃度的五加芪水煎液混合培養(yǎng)48 h后,采用MTT法檢測(cè)淋巴細(xì)胞增殖情況。T淋巴細(xì)胞分泌的IL-2和IFN-γ的濃度是通過(guò)ELISA試劑盒來(lái)進(jìn)行檢測(cè)的。體內(nèi)試驗(yàn):將7周齡的昆明系雌性小鼠進(jìn)行隨機(jī)分組,10只/組。試驗(yàn)組分為正常小鼠組(包括:正常對(duì)照組、高、中、低劑量組)和免疫抑制小鼠組(包括:正常對(duì)照組、Hy+蒸餾水組、Hy+高、中、低劑量組),連續(xù)灌胃7 d后,測(cè)定各組小鼠的免疫器官指數(shù)、碳粒廓清指數(shù)、血清溶血素水平以及脾內(nèi)抗體形成細(xì)胞(AFC)的變化。將雛雞隨機(jī)分為4組,A-D組,50只/組。A-D組在6日齡和20日齡分別免疫新城疫疫苗,B-D組在首次免疫前兩天分別灌服高、中、低劑量的中藥,每只0.5 m L,A組每只灌服0.5 m L蒸餾水,連續(xù)7 d,測(cè)定雛雞免疫器官指數(shù)、新城疫抗體水平、巨噬細(xì)胞吞噬活性、腸道Ig A分泌細(xì)胞的變化。試驗(yàn)結(jié)果表明,五加芪水煎液?jiǎn)为?dú)刺激能顯著促進(jìn)T、B淋巴細(xì)胞增殖,并且4倍稀釋組的五加芪水煎液能顯著促進(jìn)T淋巴細(xì)胞分泌IL-2,原液組五加芪水煎液能顯著促進(jìn)T淋巴細(xì)胞分泌IFN-γ(P0.01)。五加芪水煎液協(xié)同PHA能顯著促進(jìn)T淋巴細(xì)胞增殖,五加芪水煎液4倍稀釋后能夠促進(jìn)T淋巴細(xì)胞分泌的IL-2,使其含量顯著高于細(xì)胞對(duì)照組與PHA對(duì)照組,五加芪水煎液2倍稀釋后能夠顯著提高T淋巴細(xì)胞分泌的IFN-γ含量,與細(xì)胞對(duì)照組與PHA對(duì)照組相比有顯著差異(P0.01)。五加芪水煎液協(xié)同LPS可以促進(jìn)B淋巴細(xì)胞增殖,與對(duì)照組有顯著差異(P0.01)。高、中劑量五加芪水煎液使正常小鼠脾指數(shù)顯著地升高(P0.01),3個(gè)劑量的五加芪水煎液均能使正常小鼠胸腺指數(shù)和碳粒廓清指數(shù)增加明顯(P0.01或P0.05),正常高劑量組小鼠血清溶血素水平明顯高于正常對(duì)照組而且存在顯著差異(P0.01),3個(gè)劑量組脾內(nèi)抗體形成細(xì)胞數(shù)量與正常對(duì)照組相比差異不明顯(P0.05)。免疫抑制小鼠組中各中藥水煎液組小鼠的免疫器官指數(shù)、碳粒廓清指數(shù)、血清溶血素水平和脾內(nèi)抗體形成細(xì)胞數(shù)量均明顯高于Hy+蒸餾水組(P0.01或P0.05),達(dá)到正常對(duì)照組水平(P0.05),說(shuō)明五加芪水煎液使免疫抑制小鼠的免疫功能恢復(fù)至正常水平。15日齡時(shí),高、中劑量組脾指數(shù)顯著高于對(duì)照組(P0.05),各五加芪水煎液組胸腺指數(shù)和法氏囊指數(shù)均明顯地升高,與對(duì)照組有顯著差異(P0.01或P0.05)。23日齡,各劑量五加芪水煎液均能顯著增加雛雞脾指數(shù)和胸腺指數(shù),并且顯著差異于對(duì)照組(P0.01或P0.05),高、中劑量組雛雞法氏囊指數(shù)顯著高于對(duì)照組(P0.05)。31日齡,各劑量的五加芪水煎液能明顯升高雛雞的脾指數(shù)(P0.01或P0.05);而各劑量五加芪水煎液組胸腺指數(shù)和法氏囊指數(shù)與對(duì)照組無(wú)顯著差異(P0.05)。15日齡,3個(gè)濃度的五加芪水煎液均能使雛雞抗體水平顯著升高(P0.01或P0.05);23和31日齡,只有高劑量組雛雞的ND抗體水平顯著高于對(duì)照組(P0.05)。16日齡時(shí),3個(gè)劑量組吞噬指數(shù)顯著高于對(duì)照組(P0.01);在24日齡時(shí),各濃度組吞噬指數(shù)與對(duì)照組差異不顯著(P0.05)。在15、23以及31日齡時(shí),各用藥組十二指腸和空腸中Ig A分泌細(xì)胞數(shù)量均多于對(duì)照組,有顯著差異(P0.01或P0.05)。五加芪水煎液能夠提高小鼠和雛雞的免疫功能。
[Abstract]:The purpose of this experiment was to study the effect of Wujia Qi Decoction on immune function of mice and chicks. The experiment was made up of two parts in vitro and in vivo. In vitro test, the spleen lymphocytes of mice were mixed with different concentration of Wujia Qi water decoction for 48 h, and the.T lymphocyte secretion was detected by MTT method. The concentration of IL-2 and IFN- gamma was detected by the ELISA kit. In vivo test: 7 weeks old Kunming female mice were randomly divided into 10 groups. The test group was divided into normal mice group (including normal control, high, middle, low dose group) and immunosuppressive mice group (including: normal control, Hy+ distilled water group, Hy+ high, medium, low dose. After 7 d continuous gavage, the immune organ index, carbon clearance index, serum hemolysin level, and intravenic antibody forming cell (AFC) were measured in each group. The chickens were randomly divided into 4 groups, A-D group and 50 group.A-D groups at 6 and 20 days of age, respectively, and the B-D group was given high level in the first two days before the first immunization. The low dose of traditional Chinese medicine, each 0.5 m L, group A each filled with 0.5 m L distilled water and 7 d continuously, measured the immune organ index, the antibody level of Newcastle disease, the macrophage phagocytosis activity, and the intestinal Ig A secretory cells. The experimental results showed that the individual stimulation of Wujia Qi Decoction could significantly promote T, B lymphocyte proliferation, and five of the 4 times dilution group. The decoction of Astragalus can significantly promote the secretion of IL-2 in T lymphocytes. The decoction of Wujia Qi in the original solution can promote the secretion of IFN- gamma (P0.01) in T lymphocytes. The synergistic effect of Wujia Qi Decoction with PHA can significantly promote the proliferation of T lymphocytes. After 4 times dilution of Wujia Qi water decoction, the secretion of IL-2 in T lymphocytes can be promoted, and the content of the decoction can be significantly higher than that of the cell control. Group and PHA control group, after 2 times dilution of Wujia Qi water decoction can significantly increase the content of IFN- gamma secreted by T lymphocyte, and there is significant difference between the control group and the PHA control group (P0.01). The synergistic effect of Wujia Qi Decoction with LPS can promote the proliferation of B lymphocyte, and there is a significant difference between the control group and the control group (P0.01). The medium dose of Wujia Qi water decoction is normal. The spleen index of mice increased significantly (P0.01). The 3 doses of Wujia Qi Decoction could increase the thymus index and carbon particle clearance index in normal mice (P0.01 or P0.05). The serum hemolysin level of the normal high dose group was significantly higher than that of the normal control group (P0.01), and the number of antibody forming cells in the spleen in the 3 dosage groups was significantly different. The immune organ index, carbon clearance index, serum hemolysin level and the number of antibody forming cells in the spleen were significantly higher than that of the Hy+ distilled water group (P0.01 or P0.05) in the mice of the immunosuppressed group, which was significantly higher than that in the normal control group (P0.05), and reached the normal control level (P0.05), indicating the decoction of Wujia Qi. When the immune function of the immunosuppressed mice was restored to the normal level of.15 days of age, the spleen index of the medium dose group was significantly higher than that of the control group (P0.05). The thymus index and the bursa index of the Wujia Decoction group were significantly increased, and there was significant difference from the control group (P0.01 or P0.05).23 days of age, and all doses of Wujia Qi water decoction could significantly increase the embryo. The chicken spleen index and thymus index were significantly different from those in the control group (P0.01 or P0.05), and the fowl bursa index in the middle dose group was significantly higher than that of the control group (P0.05).31 days old. The spleen index (P0.01 or P0.05) of the chicks could be significantly increased in each dose of Wujia Qi water decoction (P0.01 or P0.05).
