【摘要】：對(duì)寧夏地區(qū)2011年分離到的5株H1N1亞型豬流感病毒進(jìn)行了基因組測(cè)定和遺傳進(jìn)化分析。結(jié)果顯示:寧夏地區(qū)H1N1亞型豬流感病毒具有多樣性,5株分離株可分為3類(lèi),其中1株為類(lèi)人H1N1豬流感病毒,2株為經(jīng)典豬流感病毒,另外2株為重組豬流感病毒,其PB2、PB1、PA、NP、NS基因來(lái)源于北美三元重組豬流感病毒,HA、NA、M基因來(lái)源于經(jīng)典豬流感病毒;HA蛋白裂解位點(diǎn)附近氨基酸分析顯示5株H1N1亞型豬流感分離株均為低致病性毒株;序列分析結(jié)果顯示5株病毒在HA蛋白受體結(jié)合位點(diǎn)、抗原位點(diǎn),以及NA蛋白潛在糖基化位點(diǎn)處氨基酸存在差異,這些差異具有分支特異性。5株病毒在NA和M2蛋白上均未出現(xiàn)與神經(jīng)氨酸酶抑制劑和金剛烷胺耐藥性相關(guān)的氨基酸變異,但其中3株病毒的PB2蛋白基因具有哺乳動(dòng)物適應(yīng)性變異E627K。
[Abstract]:Five strains of H1N1 subtype swine influenza virus isolated in Ningxia region in 2011 were detected by genomic analysis and genetic evolution analysis. The results showed that five strains of H1N1 subtype swine influenza virus in Ningxia region could be divided into 3 groups, one of which was human H1N1 swine influenza virus, two were classical swine influenza virus, the other two were recombinant swine influenza virus. The gene of PB2, PB1, PAP, NPNs was derived from the recombinant swine influenza virus in North America. The amino acid analysis near the cleavage site of HA protein of the classical swine influenza virus showed that the five isolates of H1N1 subtype swine flu were all low pathogenic strains. The results of sequence analysis showed that there were differences in amino acids at HA receptor binding sites, antigen sites and potential glycosylation sites of na protein. There were no amino acid variations related to neuraminidase inhibitors and amantadine resistance in na and M2 proteins, but the PB2 protein genes of the three strains had mammalian adaptive variation E627K.
