馬鏈球菌馬亞種新疆分離株菌體蛋白的粗提取及免疫效果初步評估

【摘要】:馬腺疫是由馬鏈球菌馬亞種引起的嚴(yán)重危害馬屬動物傳染病之一,該病給馬養(yǎng)殖業(yè)造成嚴(yán)重經(jīng)濟損失。可引起馬高燒,下頜淋巴結(jié)腫大等癥狀。馬鏈球菌馬亞種屬于Lancefield C型鏈球菌,該菌具有多種菌體蛋白,成為其重要的毒力因子和保護性抗原。本研究主要對馬鏈球菌馬亞種新疆分離株XZ1菌體蛋白的免疫原性與免疫保護力進行分析。本研究提取了馬鏈球菌馬亞種新疆分離株XZ1的菌體蛋白,利用SDS-PAGE方法檢測提取的菌體蛋白,分別將提取的菌體蛋白與兔抗全菌的抗體及康復(fù)馬血清反應(yīng),利用Western-blot和ELISA試驗分析其免疫原性,二者的實驗結(jié)果均表明提取的菌體蛋白具有良好的抗體識別能力且能夠與康復(fù)馬抗體反應(yīng)的能力。為探索馬鏈球菌馬亞種新疆分離株提取的菌體蛋白免疫效果,本研究利用提取馬鏈球菌馬亞種菌體蛋白免疫小鼠并檢測免疫后的抗體水平及免疫保護力。結(jié)果表明,提取的菌體蛋白免疫小鼠后,可誘導(dǎo)小鼠產(chǎn)生特異性抗體,免疫后的抗體效價高于滅活的全菌,對小鼠的免疫保護率可達到67%;表明該菌體蛋白具有良好的免疫保護效果。為進一步檢測菌體蛋白對馬的免疫效果,我們將提取馬鏈球菌馬亞種新疆分離株的菌體蛋白免疫馬并檢測免疫后抗體水平和細(xì)胞免疫水平。結(jié)果表明,該菌體蛋白能夠誘導(dǎo)馬產(chǎn)生特異性抗體并可誘導(dǎo)機體的細(xì)胞免疫應(yīng)答,二免后抗體水平及細(xì)胞免疫水平同對照組相比差異顯著(P0.01),該結(jié)果為進一步開發(fā)有效的馬腺疫的防控方法提供了理論和實踐基礎(chǔ)。
[Abstract]:The equine adenosis is one of the serious infectious diseases of the genus equine caused by Maya species of Streptococcus equine, which causes serious economic losses to the horse breeding industry. Can cause equine high fever, mandibular lymph node enlargement and other symptoms. Streptococcus equi Maya belongs to Lancefield C Streptococcus. It has many bacterial proteins and has become an important virulence factor and protective antigen. In this study, the immunogenicity and immunological protection of XZ1 bacterial protein of Streptococcus equi Maya strain from Xinjiang were analyzed. In this study, the bacterial proteins of Streptococcus equina Maya strain XZ1 were extracted. The extracted bacterial proteins were detected by SDS-PAGE method, and reacted with rabbit anti-bacterial antibody and rehabilitative horse serum, respectively. Western-blot and ELISA tests were used to analyze their immunogenicity. The results of both experiments showed that the extracted bacterial protein had good ability of antibody recognition and could react with the antibody of rehabilitative horse. In order to investigate the immunological effect of Streptococcus equina Maya isolated from Xinjiang strain, this study was conducted to immunize mice with Streptococcus equina Maya strain bacterial protein and to detect the antibody level and immune protection ability after immunization. The results showed that the specific antibody was induced by immunizing mice with the extracted bacterial protein, and the titer of the antibody after immunization was higher than that of the inactivated whole bacteria. The immune protection rate of mice was 67%, which indicated that the bacterial protein had a good immune protection effect. In order to further test the immune effect of bacterial protein on horse, we will extract the bacterial protein from Streptococcus equine Maya strain Xinjiang to immunize the horse, and detect the antibody level and cellular immunity level after immunization. The results showed that the bacterial protein could induce the production of specific antibodies and the cellular immune response of the body. Compared with the control group, the level of antibody and cellular immunity after the second immunization were significantly different (P0.01). The results provided a theoretical and practical basis for the further development of effective methods for the prevention and control of equine adenosis.
