添加通路阻斷劑對(duì)IGF-1影響奶牛乳腺上皮細(xì)胞增殖與凋亡的研究

【摘要】:試驗(yàn)旨在研究胰島素樣生長(zhǎng)因子-1(insulin-like growth factor 1,IGF-1)對(duì)奶牛乳腺上皮細(xì)胞(bovine mammary epithelial cells,BMECs)增殖的影響,為后期利用IGF-1調(diào)控乳腺發(fā)育奠定理論基礎(chǔ)。以奶牛乳腺上皮細(xì)胞為材料,首先分4組分別外源添加0(對(duì)照組)、10、50、100μg/mL IGF-1且分別培養(yǎng)12、24、48、72h,測(cè)定抑制BMECs凋亡率的最佳濃度;然后分6組:單純BMECs組、BMECs+IGF-1組、BMECs+LY294002組、BMECs+IGF-1+LY294002組、BMECs+RAPA組和BMECs+IGF-1+RAPA組,采用流式細(xì)胞術(shù)測(cè)定各組細(xì)胞凋亡率。結(jié)果表明,外源性添加IGF-1對(duì)BMECs凋亡率具有抑制作用,最佳濃度為50μg/mL;BMECs+IGF-1+LY294002與BMECs+IGF-1+RAPA組細(xì)胞凋亡率均極顯著高于BMECs+IGF-1組(P0.01)。推斷IGF-1能夠活化PI3K/Akt/mTOR信號(hào)通路,參與BMECs凋亡的調(diào)控作用,進(jìn)而抑制BMECs細(xì)胞凋亡;IGF-1可能會(huì)對(duì)被抑制PI3K/Akt/mTOR信號(hào)通路產(chǎn)生"修復(fù)"機(jī)制,使其能夠重新參與BMECs的生命進(jìn)程。
[Abstract]:The aim of the experiment was to study the effect of insulin-like growth factor 1 IGF-1 on the proliferation of bovine breast epithelial (bovine mammary epithelial cells BMECs, and to lay a theoretical foundation for the regulation of breast development by IGF-1. Dairy mammary epithelial cells were first divided into 4 groups: 10 050 渭 g / mL IGF-1 supplemented with 0 (control) 1050g / mL IGF-1 and cultured for 72 h, respectively. The optimal concentration of inhibiting apoptosis rate of BMECs was determined. Then it was divided into six groups: BMECs IGF-1 group, BMECs LY294002 group, BMECs IGF-1 LY294002 group and BMECs IGF-1 RAPA group. The results showed that exogenous addition of IGF-1 inhibited the apoptosis rate of BMECs. The best concentration of IGF-1 IGF-1 LY294002 and BMECs IGF-1 RAPA group was significantly higher than that of BMECs IGF-1 group (P0.01). It is inferred that IGF-1 can activate the PI3K / Akt / mTOR signaling pathway, participate in the regulation of BMECs apoptosis, and then inhibit the apoptosis of BMECs cells. IGF-1 may produce a "repair" mechanism on the inhibited PI3K / Akt / mTOR signaling pathway and enable IGF-1 to re-participate in the life process of BMECs.
